摘要
为提高玉米须多糖提取率并探究其降糖和抑菌活性,采用纤维素酶酶解得到玉米须多糖粗提物,然后回流提取,多糖提取率为评价指标,进行料液比、回流时间、回流温度单因素试验,采用响应面法优化玉米须多糖酶解-回流工艺条件,并测定玉米须多糖对α-葡萄糖苷酶抑制作用,对大肠杆菌、金黄色葡萄球菌和谷草芽孢杆菌的抑制作用。得到最优条件为料液比1∶30、回流时间2 h、回流温度90℃时,玉米须多糖提取率达(35.14±0.02)%。玉米须多糖能够抑制α-葡萄糖苷酶活性,抑制大肠杆菌、金黄色葡萄球菌和谷草芽孢杆菌的生长。因此玉米须多糖具有降糖作用及抑菌作用,可为玉米须多糖的开发利用提供参考。
In order to improve the extraction rate of corn silk polysaccharides and explore its hypoglycemic and antibacterial activities,cellulose extracts were used to obtain crude extracts of corn silk polysaccharides,and then extracted by reflux.The extraction rate of polysaccharides was evaluated,and the ratio of material to liquid,reflux time and reflux temperature were calculated.The single factor test of temperature was used to optimize the enzymatic hydrolysis-reflow process conditions of corn silk polysaccharides by response surface methodology,and the inhibition ofα-glucosidase by corn silk polysaccharides was determined,and the inhibition of Escherichia coli,Staphylococcus aureus and Bacillus licheniformis were determined.The optimum conditions were 1:30,reflux time 2h,reflux temperature 90°C,the extraction rate of corn silk polysaccharides was(35.14±0.02)%.Corn silk polysaccharides could inhibitα-glucosidase activity and inhibit the growth of Escherichia coli,Staphylococcus aureus and Bacillus licheniformis.Therefore,corn silk polysaccharides had hypoglycemic and antibacterial effects,which could provide reference for the development and utilization of corn silk polysaccharides.
