lncRNA AB07361在冠状动脉粥样硬化性心脏病患者血清中的表达及意义 被引量:4

摘要 目的探讨长链非编码RNA(lncRNA)AB07361在冠状动脉粥样硬化性心脏病(CAD)患者血清中的表达及其临床意义。方法选取2017年3月~2019年6月于甘肃省心血管病研究所住院的CAD患者及非CAD患者,各96例。qRT-PCR法测定两组患者外周血中lncRNA AB07361的表达及9p21基因区域3种单核苷酸(rs1333049、rs564398、rs2811712)、CDKN2A、CDKN2B的相对表达量,同时分析各变量间的相关性,通过受试者工作特征(ROC)曲线分析lncRNA AB07361的表达在CAD诊断中的敏感度与特异度。结果 qRT-PCR结果表明,在CAD患者外周血中lncRNA AB07361的相对表达量显著高于非CAD患者(P<0.05);ROC曲线结果显示,当Cut-off值为3.026,曲线下面积(AUC)值为0.883时,诊断CAD的特异度为85%,灵敏度为75%。在两组受试者中rs1333049、rs2811712及rs564398等位基因频率差异均无统计学意义(均P>0.05)。CAD组lncRNA AB07361(外显子17-18)、AB07361(外显子1-2)与rs133049等位基因G频率均呈正相关(r=0.341,0.313,均P<0.05)。CAD患者lncRNA AB07361(外显子17-18)表达与CDKN2A表达呈正相关(r=0.548,P<0.01),而与CDKN2B的表达无相关性;lncRNA AB07361(外显子1-2)与CDKN2A、CDKN2B表达均呈正相关(r=0.652,0.518,均P<0.05)。结论 lncRNA AB07361可能通过调控CDKN2B的表达而影响CAD发病与进程,而lncRNA AB07361的表达受染色体9p21区域单核苷酸rs1333049表达的影响。CAD患者外周血中lncRNA AB07361水平升高,lncRNA AB07361可作为诊断CAD的独立分子标志物。 Objective To investigate the expression of lncRNA AB07361 in serum of patients with coronary atherosclerotic heart disease(CAD)and its clinical significance,in order to provide molecular theoretical basis for clinic.Methods A total of 96 patients with CAD who were admitted to our Institute of Cardiovascular Diseases from March 2017 to June 2019 and 96 patients without CAD were enrolled.The expression of lncRNA AB07361 in blood and the relative expression of three single nucleotides(rs1333049,rs564398,rs2811712)and CDKN2 A,CDKN2 B in 9 p21 gene region were detected by qRT-PCR,and the correlation between each variable were analyzed.Sensitivity and specificity of lncRNA AB07361 expression in diagnosing CAD was analyzed.At the same time,the receiver operating characteristic(ROC)curve was plotted for the expression value of lncRNA AB07361 in the above plasma,and the area under the ROC curve(AUC)was calculated.Results The results of qRT-PCR showed that the relative expression of lncRNA AB07361 in the plasma of CAD patients was significantly higher than that of non-CAD patients(P<0.05).The ROC curve showed that when Cut-off value was 3.026 and AUC value was 0.883,the specificity and sensitivity of CAD diagnosis was 85% and 75%,respectively.There were no significant differences in the allele frequencies of rs1333049,rs281712 and rs564398 between the two groups(P>0.05).The lncRNA AB07361(exons 17-18)and lncRNA AB07361(exons 1-2)were positively correlated with the G-frequency of rs133049 allele in CAD group(r=0.341,0.313,both P<0.05).The expression of lncRNA AB07361(exons 17-18)in the CAD group was positively correlated with the expression of CDKN2 A(r=0.548,P<0.01),but not correlated with the expression of CDKN2 B.lncRNA AB07361(exons 1-2)was positively correlated with expression of both CDKN2 A and CDKN2 B(r=0.652,0.518,both P<0.05).
