摘要
研究亚麻籽木酚素对实验性高血脂症大鼠的降脂作用和抗氧化应激作用。48只大鼠随机分为6组,分别为正常组、模型组、阳性对照组、亚麻籽木酚素高、中、低剂量组(0.6、0.3、0.15 g/kg)。正常对照组给予普通饲料喂养,其余各组给予高脂饲料喂养以建立高血脂症模型。造模成功后分别灌胃给药6周。测定大鼠体重、肝肾指数,检测大鼠血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)和高密度脂蛋白(HDL-C)水平,检测大鼠肝和肾组织中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和丙二醛(MDA)的含量。肝脏脂肪明显变性。结果显示,与正常组相比,模型组大鼠的体重和肝肾指数显著增加,血清HDL-C降低,TC、TG和LDL-C升高,HE染色可见明显脂肪变性,肝肾组织中SOD和GSH-Px活性显著降低,肝组织中MDA含量显著增加。与模型组相比,亚麻籽木酚素能显著抑制高脂饮食引起的大鼠体重增加,降低肝肾指数;显著提高血清HDL-C的含量,明显降低血清中TC、TG和LDL-C的含量;抗氧化指标SOD和GSH-Px在肝肾组织中的活性显著提高,肝组织中MDA含量降低。亚麻籽木酚素低中高剂量组不同程度上减轻了大鼠肝细胞的脂肪变性情况。亚麻籽木酚素对高血脂症大鼠具有降血脂作用,其可能与降低抗氧化应激和减轻肝细胞脂肪变性有关。
To evaluate the antilipemic effect and antioxidant stress effect from flaxseed lignan in experimental hyperlipidemia rats,forty eight rats were randomly divided into normal group,model group,positive control group,high dose group(0.6 g/kg),medium dose group(0.3 g/kg)and low dose group(0.15 g/kg).The normal group was fed with ordinary feed and the other groups were fed with high-fat feed to establish hyperlipidemia model.All rats were conducted the gastrointestinal administration for 6 weeks,the body weight,liver and kidney index were calculated.The total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)in serum were detected.The content of glutathione peroxidase(GSH-Px),superoxide Dismutase(SOD)and malondialdehyde(MD)was detected in the rat livers and nephridial tissue.The results showed that compared with the normal group,weight gain,liver and kidney index significantly increased in the model group,the levels of serum HDL-C decreased,TC,TG and LDL-C increased,and HE staining showed obvious fatty degeneration.The activities of SOD and GSH-Px were also significantly decreased in liver and kidney tissue,while the MDA content was increased in liver.Compared with the model group,flaxseed lignin significantly reduced the weight gain,liver and kidney index induced by high-fat diet.The flaxseed lignin significantly elevated level of HDL-C and reduced level of TC,TG and LDL-C.The SOD and GSH-Px content were significantly increased elevated,and the MDA content was significantly reduced.The flaxseed lignin in high,medium and low dose relieved fatty degeneration.These results showed that the extract lignan from flaxseed had a lipid lowering effect on hyperlipidemia rats,which may be related to the decrement of antioxidant stress and alleviation of liver cells fat degeneration.
