摘要
目的:深入探明绵羊ERβ基因结构及其他结构与功能。方法:基于GenBank数据库中绵羊ERβ基因蛋白质编码区(CDS)区序列,利用ORFfinder、ProtParam、BioEdit、SignalP、PSORT Ⅱ、PredictProtein和SWISS MODEL等工具分析预测ERβ基因编码蛋白的结构、性质、功能及信号通路等,应用NCBI-BLAST、MEGA-X进行基因序列比对,并构建系统进化树。结果:绵羊ERβ基因CDS区共编码527个氨基酸,ERβ基因编码的产物呈疏水性,无信号肽与跨膜结构域。二级结构以α螺旋和无规卷曲为主,三级结构以α-螺旋结构单元为主。ERβ基因主要在细胞核和细胞质发挥生物学作用,这与G蛋白偶联受体活性有密切相关作用。结论:ERβ基因参与多个生殖和肿瘤相关的信号通路。
Objective The present study aimed to explore the structure and function of sheep ERβ gene and its coding products.Method The ORF finder,ProtParam,Bioedit,SignalP,PSORT Ⅱ Tools(including Predict Protein and SWISS MODEL)were applied to analyze and predict the structure,properties and functions as well as signaling pathways of ERβ gene-encoded proteins on the basis of the coding region(CDS)sequence of ovine ERβ gene in GenBank.The sequences were blasted with by NCBI-BLAST and MEGA-X,and the phylogenetic trees were constructed.Results CDs of ovine ERβ gene can encode 527 amino acids,and encoded products of ERβ gene were hydrophobicity without the signal peptide and transmembrane domain.The secondary structure containsα-helix and random coil.The tertiary structure consists ofα-helical structural units.The encoded products of ERβ gene mainly exert the biological functions in the nucleus and cytoplasm.These were greatly related to the activity of G protein coupled receptors.Conclusions The encoded products of ERβ gene were involved in many signaling pathways related to the reproduction and tumor.
