摘要
莱氏绿僵菌Metarhiziumrileyi是重要的昆虫病原真菌,对鳞翅目害虫有很好的防治效果,但存在杀虫速度慢的缺点。为了提高莱氏绿僵菌孢子的萌发速度,缩短对寄主的致死时间,在孢子悬液中分别添加不同糖类和维生素类,以测定外源营养对莱氏绿僵菌孢子萌发的促进作用。配制含有不同浓度糖和维生素的SMY液体培养基后,接种M.rileyi孢子,25℃、180r/min振荡培养,每隔24h,用血球计数板镜检孢子萌发率。结果表明:接种24h后,以10g/L甘露糖、5g/L维生素B6和5g/L维生素C对孢子萌发的促进作用较好,孢子萌发率分别为3.50%、3.67%和0.83%;随着培养时间延长,10g/L甘露糖、5g/L维生素C依然对孢子萌发有较好的促进作用,5g/L维生素B6对孢子萌发的促进作用逐渐降低;进一步测定甘露糖和维生素C对孢子萌发的浓度效应,甘露糖浓度为25.0g/L时对孢子萌发的促进作用最好,维生素C浓度为0.2g/L时最好。以上结果表明,对M.rileyi孢子萌发促进效果最好的外源营养是甘露糖和维生素C,最适浓度分别是25.0 g/L和0.2g/L。
Metarhizium rileyi is an important entomopathogenic fungus with good control effect against Lepidopteran pests,while its insecticidal speed was slow.Hence,the promotion effects of external saccharides and vitamins on spore germination of Metarhizium rileyi were analyzed to find a way to shorten the lethal time and improve the spore germination speed of M.rileyi on host.The conidia of M.rileyi were inoculated into 6 saccharide and 4 vitamin fluid media,and cultured in the conditions of 25℃and 180 r/min on the incubator shaker;the germinated spores were counted by hemocytometer under microscopy every 24 h after inoculation.The results showed that 10 g/L mannose,5 g/L vitamin B6 and 5 g/L vitamin C had better promotion effects on spore germination after 24 h,and the spore germination rates were 3.5%,3.67%and 0.83%respectively.With the extension of culture time,10 g/L mannose and 5 g/L vitamin C still had better promoting effects on spore germination,while the promoting effect of 5 g/L vitamin B6 gradually decreased.Further study showed that mannose had the best promotion effect when the concentration was 25.0 g/L,while the optimum concentration of vitamin C was 0.2 g/L.It was concluded that mannose and vitamin C were the best external nutrients in promoting spore germination of M.rileyi at the concentrations of 25.0 g/L and 0.2 g/L,respectively.
