摘要
以海带渣为原料,分别研究稀酸-黑曲霉和稀酸-纤维素酶2种酶的糖化效果,确定了较优的糖化工艺参数。结果表明,稀酸-黑曲霉糖化海带渣的最佳条件为接种量10%,发酵温度32℃,pH5.2,发酵时间60 h时,还原糖产率为29.37%;稀酸-纤维素酶糖化海带渣的最佳条件为酶用量600 U/g,酶解温度52℃,pH5.2,酶解时间36 h时,还原糖产率为32.17%。2种糖化方法相比,稀酸-纤维素酶的还原糖产率略高,但从节约成本考虑,较优的糖化方法为稀酸-黑曲霉糖化法。
With Laminaria japonica residue as raw materials, the effects of saccharification by the dilute acid-aspergillus niger method and dilute acid-cellulase method were studied. The optimum saccharification parameters were obtained. The results showed that the best conditions of the dilute acid-Aspergillus niger saccharification were inoculation amount 10 %, fermentation temperature 32 ℃, pH 5.2, fermentation time 60 h.Under the optimized conditions, the reducing sugar yield rate was 29.37 %;The best conditions of the dilute acid-cellulase saccharification were cellulase dosage 600 U/g, temperature 52 ℃, pH 5.2, hydrolysis time 36 h. Under the optimized conditions, the reducing sugar yield rate was 32.17 %. Compared with the two saccharification methods, the reducing sugar yield of dilute acid-cellulase is slightly higher. But based on the cost saving, the better method of saccharification is dilute acid-Aspergillus niger method.
