摘要
【目的】海洋微生物是海洋天然产物和药物研究的宝贵资源,已有大量结构新颖、活性显著的化合物从微生物中被发现。由于海洋真菌遗传背景复杂、代谢产物种类丰富,故其已成为发掘新天然产物的重要资源,从海洋生物中寻找新的天然抗氧化活性物质成为目标之一。【方法】从海南省西沙群岛附近海域采集的聚裂丛柳珊瑚(Rumphella aggregata)和华丽肉芝软珊瑚(Sarcophyton elegans)中分离纯化得到13株共附生真菌,并通过测定对高价铁离子的还原作用、DPPH自由基的清除能力、总酚和总黄酮的含量、ABTS自由基的清除能力、羟基自由基的清除能力来对13株真菌发酵液进行抗氧化能力进行评估。【结果】测定结果表明,13株真菌发酵液均具有较好的抗氧化活性,其中Z1菌株对DPPH和羟基自由基的清除率最高,分别达到78.03%和73.36%;S9菌株对高价铁离子的还原能力和对ABTS自由基的清除能力最强,分别达到了130.56 mmol/L Fe2+/g DW和84.09%。通过对Z1和S9菌株的形态学特征观察和18S rDNA序列进行分析,初步将菌株Z1鉴定为黑曲霉菌Aspergillus niger,菌株S9鉴定为Paraconiothyrium brasiliense。【结论】黑曲霉菌Aspergillus niger和Paraconiothyrium brasiliense的发酵液具有良好的抗氧化活性,可对这两株菌株进行下一步的活性物质追踪研究。
[Objective]Marine microorganisms are valuable resources for the research of marine natural products and drugs.A large number of compounds with novel structure and remarkable activity have been found from microorganisms.Because of the complex genetic background and rich types of metabolites,marine fungi have become an important resource for exploring new natural products,so looking for new natural antioxidant substances from marine organisms has become one of the targets.[Method]Thirteen strains of symbiotic fungi were isolated and purified from Rumphella aggregata and Sarcophyton eleganscollected from Xisha Islands in Hainan Province.To evaluate antioxidant ability,the reducing power of high valence iron ion,the content of total phenols and flavonoids and the scavenging ability of DPPH,ABTS and hydroxyl was determined.[Result]The results showed that the scavenging rates of DPPH and hydroxyl radicals of strain Z1 were 78.03%and 73.36%,respectively.Strain S9 had the strongest ability to reduce high valence iron ions and scavenging ABTS free radi⁃cals,reaching 130.56 mmol/L Fe2+/g DW and 84.09%,respectively.Through the analysis of the morphological characteristics and 18s rDNA sequence of Z1 and S9 strains,Z1 strain was identified as Nigrospora oryzae,S9 strain was Paraconiothyrium brasiliense.[Conclusion]The fermentation broth of Aspergillus niger and Paraco⁃niothyrium brasiliense have good antioxidant activity,so the active substances of these two strains can be traced in the next step.
