积雪草苷对烫伤大鼠创面愈合及瘢痕形成中热休克蛋白47表达的影响 被引量:13

摘要 目的探讨积雪草苷对烫伤大鼠创面愈合及瘢痕形成中热休克蛋白47(HSP47)表达的影响。方法对24只SD大鼠进行深Ⅱ度烫伤建模;采取同体对照处理,右侧创面外敷积雪草苷(积雪草苷组),外覆盖凡士林油纱;左侧创面直接覆盖凡士林油纱(对照组)。烫伤后第3、7、14、21天,每次随机取6只大鼠,评估创面愈合情况;取左右侧创面标本,免疫组化染色法检测HSP47表达量。结果烫伤后观察大鼠21d,积雪草苷组创面全部愈合,对照组创面仅4个愈合。大鼠烫伤后第7、14天,积雪草苷组创面HSP47表达量明显高于对照组(均P<0.05),第14天后开始缓慢下降,第21天两组创面HSP47表达量比较差异无统计学意义(P>0.05)。结论积雪草苷可能通过影响HSP47的表达来促进胶原成熟和创面愈合,从而减轻瘢痕形成。 Objective To investigate the expression of heat shock protein 47(HSP47)during the process of wound healing and scar formation in scalded rats treated with asiaticoside.Methods The deep second degree scald was induced in 24 SD rats.The right side of the wound was covered with asiaticoside(asiaticoside group)and vaseline gauze,the left side of the wound was covered with vaseline gauze only(control group).On the d3,d7,d14 and d21 after scald,6 rats were randomly taken in each batch,the wound healing was evaluated and the expression of HSP47 was detected by immunohistochemistry in the left and right wound samples.Results On d21 after scalding,the wounds in asiaticoside group healed completely,while wounds healed only in 4 rats in control group.On the d7 and d14 after scalding,the expression of HSP47 in the asiaticoside group was significantly higher than that in the control group(P<0.05),the HSP47 expression level started to decrease slowly afterwards,and on d21 there was no significant difference between the two groups(P>0.05).Conclusion Asiaticoside can promote wound healing with less scar,which may be associated with the expression of HSP47.
