摘要
目的研究于氏头穴丛刺结合丰富环境对APP/PS1转基因小鼠海马神经细胞凋亡的影响,探讨其相关作用机制。方法釆用随机数字表法将36只雄性7月龄APP/PS1双转基因小鼠随机分为模型组、针刺组、康复组、针康组各9只。另外9只同性别、同月龄的野生小鼠作为空白组。将康复组小鼠置于丰富环境中,针刺组给予头穴丛刺作为基础治疗,针康组小鼠在丰富环境中给予头穴丛刺治疗。针刺神庭透百会以及左右两旁2mm,15 min/矢连续治疗4周。采用免疫组化法观察海马Bax、Bcl-2蛋白表达的变化。结果与空白组比较,模型组Bcl-2阳性细胞表达相对少,而Bax阳性细胞表达相对增加(PV0.01)。与模型组比较,针刺组、康复组、针康组海马区bax表达均显著下降(P<0.01),Bcl-2阳性神经元计数明显升高(P<0.01)。与针刺组、康复组比较,针康组Bcl-2阳性神经元计数明显升高,Bax表达明显下降(P<0.05).针刺组与康复组之间比较无统计学差异(P>0.05).结论头穴丛刺结合丰富环境减轻了小鼠海马的细胞凋亡情况。
Objective To study the effect of cluster needling at Yu's scalp point combined with enriched environment on apoptosis of hippocampal neurons in APP/PS1 transgenic mice and explore its related mechanism.Methods 36 male 7-month-old APP/PS1 double transgenic mice were randomly divided into model group,acupuncture group,rehabilitation group and needle-Kang group with 9 mice in each group.Another 9 wild mice of the same sex and age were used as blank group.Rehabilitation group mice were placed in a rich environment.Acupuncture group mice were given cluster needling at scalp points as the basic treatment.Acupuncture group mice were given cluster needling at scalp points in a rich environment.Acupuncture of Shenting Toubaihui and its left and right sides for 2 mm,15 min/time for four weeks.The expression of Bax and Bcl-2 protein in hippocampus was observed by immunohistochemistry.Results Immunohistochemical results:Compared with the blank group,the expression of Bcl-2 positive cells in the model group was relatively low,while the expression of Bax positive cells was relatively increased (P < 0.01). Compared with the model group, the expression of Bax in the hippocampus ofthe acupuncture group, rehabilitation group and acupuncture-Kang group decreased significantly (P V 0.01), andthe number of Bcl-2 positive neurons increased significantly (P < 0.01). Compared with the acupuncture group andrehabilitation group, the number of Bcl-2 positive neurons was significantly increased and the expression of Bax wassignificantly decreased in the acupuncture group (P < 0.05). There was no statistical difference between acupuncturegroup and rehabilitation group (P > 0.05). Conclusion Scalp acupoint cluster needling combined with enrichedenvironment can alleviate the apoptosis of hippocampal cells in mice.
