摘要
依据黄酮类化合物可以与Al^3+形成稳定的荧光络合物,建立了一种银杏叶提取物中总黄酮含量的测定方法。以芦丁为标准样品,采用荧光光度法,在激发波长442 nm,发射波长482 nm范围内,测定银杏叶提取物中总黄酮的含量,并进行精密度和加标回收实验。在最优的条件下,荧光强度与芦丁浓度在4.00~20.0 mg/L的范围内有良好的线性关系。该方法的检出限为0.062 mg/L,平均回收率为99.4%,相对标准偏差(RSD)小于2%。该方法操作简便,检测高效快速,结果准确,数据重现性好,适合于银杏叶提取物中总黄酮含量的测定。
The method for determination of total flavonoid in Ginkgo biloba extract is established based on a stable fluorescent complex formed by the flavonoids with Al^3+.Taking rutin as the standard sample,the content of total flavonoids in Ginkgo biloba extract was determined at the excitation wavelength of 442 nm and the emission wavelength of 482 nm.The precision and accuracy of the method were also evaluated.Under the optimal conditions,the fluorescence intensity has a good linear relationship with the concentration of rutin in the range of 4.00~20.0 mg/L.The detection limit of this method is 0.062 mg/L,the average recovery was 99.4%,and the relative standard deviation(RSD)was less than 2%.The method is simple,convenient and efficient.The result is accurate and with good reproducibility.Therefore,the method is suitable for determining the total flavonoids in the extract of Ginkgo biloba leaves.
