摘要
目的:探讨黑色素瘤相关抗原A9(MAGE-A9)、MAGE-A11-和Ki67在喉鳞状细胞癌组织中的表达,并分析其与喉鳞状细胞癌患者临床病理学特征及预后的关系。方法:选取河北医科大学第四医院2012-2014年住院手术切除的喉鳞状细胞癌组织及相应癌旁组织标本各73例,同时选取该院3例前列腺癌住院患者去势治疗术后的睾丸组织作为阳性对照,应用免疫组织化学法检测喉鳞状细胞癌组织及相应癌旁组织中MAGE-A9、MAGE-A11和Ki67蛋白的表达水平。结果:喉鳞状细胞癌组织中MAGE-A9、MAGE-A11蛋白和Ki67的表达率[47.94%(35/73)]、[49.32%(36/73)]和[46.58%(34/73)]均显著高于相应的癌旁组织[0(0/75)(P<0.01)],MAGE-A9和MAGE-A11蛋白的表达与喉鳞状细胞癌患者的临床分期和淋巴转移有关(P<0.05),Ki67LI表达与喉鳞状细胞癌患者的肿瘤大小、临床分期和淋巴转移有关(P<0.05)。相关性分析显示,MAGE-A9和MAGE-A11蛋白表达均与Ki67指数呈正相关(r=0.258,P=0.027;r=0.672,P=0.001)。Kaplan-Meier生存曲线分析显示,MAGE-A9、MAGE-A11和Ki67LI蛋白高表达阳性患者的生存率均显著低于低表达的患者,而且MAGE-A9和Ki67LI均高表达或MAGE-A11和Ki67LI均高表达患者的生存期均显著低于其单一高表达或均低表达的患者(P<0.05或P<0.01)。单因素和多因素Cox回归模型分析进一步提示,MAGE-A9蛋白和MAGE-A11蛋白是喉鳞状细胞癌患者总体生存的独立预后因素(P<0.05)。结论:MAGE-A9、MAGE-A11和Ki67是喉鳞状细胞癌的肿瘤相关抗原,其可作为预测喉鳞状细胞癌患者的预后指标。
Objective:To explore the expressions of melanoma antigen(MAGE)-A9,-A11 and Ki67 in laryngeal squamous cell carcinoma(LSCC) tissues,and to analyze their correlation with clinicopathological features and the prognosisof LSCC patients.Methods:A total of 73 pairs of LSCC tissuesand corresponding para-cancerous tissues resected from LSCC patients,who were treated at the Fourth Hospital of Hebei Medical University from 2012 to 2014,were collected for this study.At the same time,testicular tissues from 3 patients with prostate cancer after castration were selected as positive control.The protein expressions of MAGE-A9,MAGE-A11 and Ki67 in LSCC tissues and its para-cancerous tissues were detected by immunohistochemistry.Results:The expression rates of MAGEA9,MAGE-A11 protein and Ki67 in LSCC tissues were 47.94%(35/73),49.32%(36/73) and 46.58%(34/73) respectively,which were significantly higher than those in para-cancerous tissues.The protein expressions of MAGE-A9 and MAGE-A11 were correlated with clinical stage and lymphatic metastasis of LSCC(P<0.05).The expression of Ki67 LI was correlated with tumor size,clinical stage and lymphatic metastasis of LSCC(P<0.05).The correlation analysis showed that the expressions of MAGE-A9 and MAGE-A11 were positively correlated with Ki67(r=0.258,P=0.027;r=0.672,P=0.001).Kaplan-Meier survival curve analysis showed that the survival rates of patients with high expression of MAGE-A9 protein(P=0.009),MAGE-A11 protein(P=0.031) and Ki67 LI(P=0.040) were significantly lower than those with low expressions.And the survival time of patients with both high expressions of MAGE-A9 and Ki67 LI(P=0.001) or both high expressions of MAGE-A11 and Ki67(P=0.001) was significantly shorter than that of patients with low expression(both or single).Univariate and multivariate Cox regression analysis further indicated that MAGE-A9 protein(P=0.028) and MAGE-A11 protein(P=0.042) were independent prognostic factors for overall survival of LSCC patients.Conclusion:MAGE-A9,MAGE-A11 and Ki67 are tumor-associated
