摘要
目的研究黑色素瘤相关抗原A9(melanoma-associated antigen-A9,MAGE-A9)表达对卵巢癌细胞增殖、克隆形成、迁移和侵袭的影响。方法对MAGE-A9高表达细胞株HO-8910、SKOV3进行MAGE-A9敲减处理,对MAGE-A9低表达细胞株A2780、OVCAR-3进行MAGE-A9过表达处理;qRT-PCR和Western Blot检测MAGE-A9 mRNA和蛋白表达情况;CCK-8实验、细胞克隆形成实验、细胞划痕愈合实验、Transwell侵袭实验分别检测干扰和过表达MAGE-A9对卵巢癌细胞增殖、克隆形成、迁移和侵袭能力的影响。结果MAGE-A9基因在HO-8910、SKOV3细胞中的表达被成功干扰,在A2780、OVCAR-3细胞中被成功过表达。相对于对照组,MAGE-A9基因过表达处理的A2780、OVCAR-3细胞的增殖、克隆形成、迁移和侵袭能力增强;MAGE-A9基因干扰处理的HO-8910、SKOV3细胞的增殖、克隆形成、迁移和侵袭能力减弱。结论MAGE-A9表达可以促进卵巢癌细胞的增殖、克隆形成、迁移和侵袭。
Objective To investigate the effects of melanoma-associated antigen-A9(MAGE-A9)gene on the proliferation,colony formation,migration and invasion abilities of ovarian cancer cells.Methods HO-8910 and SKOV3 lines which high expression MAGE-A9 were disturbanced expression of MAGE-A9,A2780 and OVCAR-3 lines which low expression MAGE-A9 were overexpressed MAGE-A9.The expression levels of MAGE-A9 mRNA and protein were detected by qRT-PCR and Western Blot.The effects of disturbancing or overexpressing MAGE-A9 on the proliferation,colony formation,migration and invasion abilities of ovarian cancer cells were investigated by cell CCK-8,colony-forming assay,wound healing assay and Transwell assay,respectively.Results The MAGE-A9 gene were successfully disturbanced in HO-8910 and SKOV3 cells,and successfully overexpressed in A2780 and OVCAR-3 cells.Compared with the control group,the proliferation,colony formation,migration and invasion abilities of A2780 and OVCAR-3 cells overexpressed with MAGE-A9 were significantly enhanced,the proliferation,colony formation,migration and invasion abilities of HO-8910 and SKOV3 cells disturbanced with MAGE-A9 were significantly inhibited.Conclusion The expression of MAGE-A9 enhances the proliferation,colony formation,migration and invasion abilities of ovarian cancer cells.
