微小RNA-92a靶向Krüppel样因子2基因促进卵巢癌细胞侵袭的作用

摘要 目的 研究微小RNA-92a(miR-92a)靶向Krüppel样因子2(KLF2)基因促进卵巢癌细胞侵袭的作用及机制。方法2019年1月至2020年3月进行本研究。培养卵巢癌细胞系A2780、SKOV3、OVCAR-3及卵巢上皮细胞系HOSEpiC,检测miR-92a的表达水平;SKOV3细胞分为阴性对照(NC)组、miR-92a抑制物组、miR-92a组、pcDNA组、pcDNA+miR-92a组、pcDNAKLF2+miR-92a组,分别转染NC序列、miR-92a、miR-92a抑制物、空白pcDNA质粒、过表达KLF2的pcDNA重组质粒,检测细胞侵袭数目、KLF2表达水平,双荧光素酶报告基因验证miR-92a靶向KLF2基因。结果 A2780、SKOV3、OVCAR-3细胞中miR-92a的表达水平分别为(0.92±0.15)、(1.62±0.19)、(1.21±0.13),均高于HOSEpiC的(0.62±0.09)(P<0.05);与NC组(75.38±13.82)、(0.42±0.08)比较,miR-92a抑制物组SKOV3细胞的侵袭数目(42.38±8.79)减少、KLF2表达水平(0.78±0.20)增加,miR-92a组SKOV3细胞侵袭数目(109.38±21.38)增加、KLF2表达水平(0.29±0.06)减少,野生型KLF2双荧光素酶报告基因的荧光活力降低(P<0.05);与pcDNA组比较、pcDNA+miR-92a组SKOV3细胞的侵袭数目增加,与pcDNA+miR-92a组比较、pcDNAKLF2+miR-92a组SKOV3细胞的侵袭数目减少(P<0.05)。结论 卵巢癌细胞中miR-92a表达增加,miR-92a能够促进SKOV3细胞侵袭、其机制可能与靶向调控KLF2基因表达有关。 Objective To study the role and mechanism of microRNA-92a(miR-92a) targeting the Krüppel-like factor 2(KLF2) gene to promote ovarian cancer cell invasion.MethodThe study was conducted from January 2019 to March 2020.Ovarian cancer cell lines A2780,SKOV3,OVCAR-3 and ovarian epithelial cell line HOSEpiC were cultured to detect the expression level of miR-92a;SKOV3cells were divided into negative control(NC) group,miR-92a inhibitor group,miR-92a group,pcDNA group,pcDNA+miR-92a group,pcDNA-KLF2+miR-92a group,transfected with NC sequence,miR-92a,miR-92a inhibitor,blank pcDNA plasmid and pcDNA recombinant plasmid overexpressing KLF2,respectively.The number of invaded cells and the expression level of KLF2 were detected,and a dual-luciferase reporter gene verified that miR-92a targeted the KLF2 gene.ResultThe expression levels of miR-92a in A2780,SKOV3and OVCAR-3 cells were 0.92±0.15,1.62±0.19 and 1.21±0.13,respectively,which were higher than those in HOSEpiC cells(0.62±0.09)(P < 0.05).Compared with the NC group(75.38±13.82),(0.42±0.08),the invasion number of SKOV3 cells in the miR-92a inhibitor group(42.38±8.79) decreased,and the expression level of KLF2 increased(0.78±0.20).In the miR-92a group,the invasion number of SKOV3 cells increased(109.38±21.38),the expression level of KLF2 decreased(0.29±0.06),and the fluorescence activity of the wildtype KLF2 dual-luciferase reporter gene decreased(P<0.05).Compared with the pcDNA group,the invasion number of SKOV3 cells in the pcDNA+miR-92a group increased,and compared with the pcDNA+miR-92a group,the invasion number of SKOV3 cells in the pcDNA-KLF2+miR-92a group decreased(P < 0.05).Conclusions miR-92a can promote the invasion of SKOV3 cells.The mechanism may be related to the targeted regulation of KLF2 gene expression.
