基于GC/TOF-MS代谢组学方法的人参反藜芦研究 被引量:4

摘要 目的利用GC/TOF-MS技术,从整体上认识人参与藜芦合用后对动物代谢的影响,并从新的角度为人参反藜芦寻找科学依据。方法将102只实验动物分为正常对照组、单煎藜芦组、单煎人参组、合并组、合煎组、前后给药组,每组17只。按照藜芦剂量2.5 g/kg,人参25 g/kg,分别灌胃给予蒸馏水、藜芦单煎液、人参单煎液、人参藜芦合并液、人参藜芦合煎液。人参藜芦前后给药组先给人参单煎液,1 h后给予藜芦单煎液。所有动物单次给药后取血清进行代谢差异分析,即代谢谱的主成分分析(PCA)得分图和偏最小二乘法判别分析(PLS-DA)得分图,其中权重系数VIP>1表示有差异代谢物。结果给药后,每组仅有10只小鼠的数据有效。代谢组学结果表明,给药组与正常组在PLS-DA模式的得分图上有差异,单煎藜芦组和合煎组、合并组和合煎组、前后给药组和合并组,三类比较在PCA、PLS-DA模式的得分图上都有显著差异,且差异代谢物明显。结论结果表明人参和藜芦的配伍确实对生物体产生了影响,为藜芦和人参配伍后内源性代谢物靶标的发现提供依据。 Objective To explore animal physiological and metabolic effects after combination of veratrum nigrum and ginseng as a whole by the GC/TOF-MS technology,and provide a scientific basis from a new angle for the effect of Ginseng in combination with veratrum nigrum.Methods The mice were randomly divided into five groups(n=17 in each group):control group,veratrum nigium group,ginseng group,mixture group,mixed-decoction group,combination group.The distilled water,2.5 g/kg veratrum nigium single decoction,25 g/kg ginseng single decoction,the mixture of ginseng and veratrum nigium decoctions,the mixed-decoction of ginseng and veratrum nigium were gavaged in control group,veratrum nigium group,ginseng group,mixture group,mixed-decoction group,respectively.In combination group,the mice were first given ginseng extract,and then given veratrol extract after 1 h.Serum was taken for metabolism difference analysis after administration.The weight coefficient of VIP>1 represents the metabolite with difference.Results Only 10 mice in each group were effective for data collection.Metabonomics results showed that the score in PLS-DA score plot from metabolic profiles was different between dosage group and normal group.PCA,PLS-DA scores plot from metabolic profiles had significant difference between veratrum nigium group and mixed-decoction group,between mixture group and mixed-decoction group,ginseng group,mixture group,between mixture group and combination group,and had different metabolites.Conclusion Compatibility application of ginseng and veratrum nigrum in modern medicine is reasonable,and provides basis for seeking the target of metabolites after compatibility of ginseng and veratrum nigrum.
