牙龈卟啉单胞菌脂多糖感染对糖尿病兔肾损害的影响及相关机制 被引量:4

摘要 目的在兔糖尿病模型基础上建立微量牙龈卟啉单胞菌(Pg)内毒素血症模型,评价Pg-LPS感染对肾损害的影响及其分子机制。方法将20只新西兰兔随机分为正常对照组、DM+ND+Pg-LPS组(糖尿病建模+普通饲料+Pg-LPS注射)、DM+HD组(糖尿病建模+高脂饲料)、DM+HD+Pg-LPS组(糖尿病建模+高脂饲料+Pg-LPS注射),每组5只。除正常对照组外,其余各组均给予四氧嘧啶(ALX)诱导糖尿病动物模型。糖尿病建模后,DM+HD组和DM+HD+Pg-LPS组喂养高脂饲料,DM+ND+Pg-LPS组及DM+HD+Pg-LPS组行微量Pg-LPS兔耳缘静脉注射。比较四组动物不同时间的空腹血糖、24 h尿量、尿蛋白(Upro)、血肌酐水平、肌酐清除率(CCr)、尿素氮(BUN)、外周血CRP、TNF-α水平、TLR4及NF-κBp65蛋白表达情况。结果建模后第1天、第4、8周末,DM+ND+Pg-LPS组、DM+HD组、DM+HD+Pg-LPS组的空腹血糖均高于正常对照组(P<0.01)。建模后第4、8周末,DM+ND+Pg-LPS组、DM+HD组、DM+HD+Pg-LPS组的24 h尿量、Upro均高于正常对照组,且DM+HD+Pg-LPS组高于DM+ND+Pg-LPS组与DM+HD组(P<0.01)。建模后第4、8周末,DM+ND+Pg-LPS组、DM+HD+Pg-LPS组的血肌酐、BUN水平高于正常对照组与DM+HD组,CCr低于正常对照组与DM+HD组,且DM+HD+Pg-LPS组的血肌酐、BUN水平高于DM+ND+Pg-LPS组,CCr低于DM+ND+Pg-LPS组(P<0.01)。建模后第4、8周末的CRP、TNF-α水平及建模后第8周末的TLR4、NF-κBp65表达水平比较,正常对照组<DM+HD组<DM+ND+Pg-LPS组<DM+HD+Pg-LPS组(P<0.01)。结论微量Pg-LPS静脉注射可以加速糖尿病兔模型的肾功能损害,其机制可能与Pg-LPS诱发兔外周血及肾脏局部的炎症免疫反应相关。 Objective To establish a model of endotoxemia of micro Porphyromonas gingivalis(Pg)based on rabbit diabetes model,to evaluate the effect of Pg-LPS infection on renal damage and its molecular mechanism.Methods Twenty New Zealand rabbits were randomly divided into normal control group,DM+ND+Pg-LPS group(diabetes modeling+normal diet+Pg-LPS injection),DM+HD group(diabetes modeling+high-fat diet),DM+HD+Pg-LPS group(diabetes modeling+high-fat diet+Pg-LPS injection),with 5 rabbits in each group.Except for the normal control group,the other groups were given alloxan(ALX)to induce diabetic animal model.After diabetic modeling,the DM+HD group and the DM+HD+Pg-LPS group were fed with high-fat diet,and the DM+ND+Pg-LPS group and DM+HD+Pg-LPS group were injected with micro Pg-LPS by ear vein.The fasting blood glucose,24-hour urine volume,urinary protein(Upro),levels of serum creatinine,creatinine clearance rate(CCr),levels of urea nitrogen(BUN),peripheral blood CRP,TNF-α,the protein expression of TLR4,NF-κBp65 were compared among the four groups.Results The fasting blood glucose of the DM+ND+Pg-LPS group,DM+HD group and DM+HD+Pg-LPS group was higher than that of the normal control group at the first day,the end of the fourth and the eighth week after modeling(P<0.01).At the end of the 4th and the 8th week after modeling,the 24-hour urine volume,Upro of the DM+ND+Pg-LPS group,DM+HD group and DM+HD+Pg-LPS group were higher than those of the normal control group,and those of the DM+HD+Pg-LPS were higher than the DM+ND+Pg-LPS group and DM+HD group(P<0.01).At the end of the 4th and the 8th week after modeling,the levels of serum creatinine and BUN of the DM+ND+Pg-LPS group and the DM+HD+Pg-LPS group were higher than those of the normal control group and the DM+HD group,the CCr was lower than that of the normal control group and the DM+HD group,the level of serum creatinine and BUN in the DM+HD+Pg-LPS group were higher than those in the DM+ND+Pg-LPS group,and CCr was lower than that in the DM+ND+Pg-LPS group(P<0.01).Compared the
