摘要
为研究肉鸡腹水综合征发病机制和中药复方的防治作用,以多因素法复制肉鸡腹水综合征模型,将304羽7日龄罗斯肉鸡随机分为对照组(常规饲养),模型组(9~11℃,饲料中添加3%猪油和4%鱼粉,饮水中添加0.12%NaCl),中药复方高、中、低剂量组(饲养同模型组,同时饮水中分别给予2、1.5、1 mL/(kg·d)中药复方),观察肉鸡心肺组织病理变化,检测心肺组织信息沉默调节因子1(SIRT1)、过氧化物酶体增殖物激活受体γ共激活因子1-α(PGC-1α)、Bcl-2、Bax蛋白含量及血清总超氧化物歧化酶(T-SOD)活性和丙二醛(MDA)含量。结果表明,35日龄时,与对照组相比,模型组肉鸡心和肺组织结构损伤、炎性细胞浸润,Bax蛋白极显著增加(P<0.01),SIRT1、PGC-1α、Bcl-2蛋白及Bcl-2/Bax比值极显著降低(P<0.01),血清T-SOD活性极显著降低(P<0.01),MDA含量极显著增加(P<0.01);与模型组相比,中药复方高、中、低剂量组肉鸡心组织细胞结构和形态均有改善,肺组织细胞结构较完整,但仍有大量炎性细胞浸润;心肺组织SIRT1、PGC-1α、Bcl-2、Bax、Bcl-2/Bax比值及血清T-SOD、MDA等指标均有不同程度改善(P<0.01或P<0.05)。SIRT1、PGC-1α及其介导的相关因子参与了肉鸡腹水综合征病理发生发展;中药复方通过调节以上活性因子,有效防治肉鸡腹水综合征。
In order te study the pathogenesis of broilere ascites syndrome(AS)and te investiaate the preventive and therapeutic effects of compound traditional Chinese medicine on AS,the broiler AS model was duplicated by multi-factor approach.Total of 3047-day-old Ross broilers wero randomly divided inte five groups.Control group was bred with basal teed.Model group was bred with mixed feed(9-11 k,added 3%lard and 4%fish meet te basal feed,and 0.12%added NaCl te drinking water).Compound traditional Chinese medicine high,medium and low dose groups were bred with mixed feed and given a dose of 2,1.5,1 mL%(kg·d)^-1 compound traditional Chinese medicine in drinking watee,respectively.Afterwards,histopathoyfical changes in heart and lung were assessed.The protein expression of SIRT1,PGC-1a,Bcl-2,Bae in heart lung,the T-SOD activity and MDA content in serum were anal ped.At 35 days of aae,compared with control group,heart and lung tissues of broiler were damaged and inflammatory cell infiltration was obsened in model group.The protein expression of Bae was significantly increased(P<0.01).The protein expression of SIRT1,PGC-1αand Bcl-2 and the ratio of Bcl-2/Bae was significantly increased(P<0.01),the activities of T-SOD in serum were significantly decreased(P<0.01)and the contents of MDA in serum were significantly increased(P<0.01).Moreover,compared with model group,Xter treatment with compound traditional Chinese medicine in high,medium and low dose groups,the heart tissues cell stwctuw and morpholovy of broiler wew improved,the lung tissues stwctuw were wlatively intact,but thew were still a lot of inflammamw ccll infiltration,the SIRT1,PGC-1 a,Bcl-2,Bae in heart and lung and the T-SOD and MDA in serum improved te some extent(P<0.01 or P<0.05).Our results showed that SIRT1,PGC-1 a and relevant factors were invelved in the pathogenesis of ascites syndrome,and compound traditional Chinese medicine could prevent and treat broilere with ascites syndromeby improving these factors.
