毛诃子提取物对高原肺动脉高压大鼠心脏的保护作用 被引量:9

摘要 目的研究毛诃子提取物对高原肺动脉高压(HAPH)所致大鼠心脏的改善作用。方法建立高原肺动脉高压大鼠,随机分为平原组(生理盐水4 mL/kg)、高原肺动脉高压组(生理盐水4 mL/kg)、阳性组(硝苯地平2.7 mg/kg)、毛诃子醇提物组(270 mg/kg)和毛诃子水提物组(270 mg/kg),连续灌胃15 d后,测定各组大鼠肺动脉血压(PAP),血清中促红细胞生成素(EPO)、C-反应蛋白(CRP)、促生长因子(IGF-1)含量,观察心脏组织病理学变化并测定其超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)水平。结果与平原组比较,高原肺动脉高压组、阳性组、毛诃子醇提物、水提物组大鼠PAP升高,血清中CRP、EPO水平均升高,心肌中SOD、GSH-Px活性减弱,MDA含量升高,差异有统计学意义(P<0.05);高原肺动脉高压组、毛诃子醇提物、水提物组大鼠血清中IGF-1含量升高,阳性组大鼠血清中IGF-1含量降低,差异有统计学意义(P<0.05)。与高原肺动脉高压组比较:阳性组、毛诃子醇提物组大鼠PAP降低,差异有统计学意义(P<0.05);阳性组、毛诃子醇提物、水提物组大鼠血清中CRP、EPO含量下降,差异有统计学意义(P<0.05);毛诃子醇提物、水提物组大鼠血清中IGF-1含量升高,心脏组织中SOD、GSH-Px活性降低,MDA含量升高,差异有统计学意义(P<0.05);阳性组大鼠血清中IGF-1含量下降,差异有统计学意义(P<0.05)。结论毛诃子醇提物和水提物对HAPH大鼠心脏损伤均有不同程度的改善作用。 Objective To investigate the improvement of the extract of Terminalia billerica Roxb.(TBR) on the rat heart induced by high altitude pulmonary hypertension(HAPH). Methods A rat model of HAPH was established and randomly divided into plain group(normal saline 4 mL/kg), HAPH model group(normal saline 4 mL/kg), positive group(nifedipine 2.7 mg/kg), alcohol extract of TBR group(270 mg/kg) and water extract of TBR group(270 mg/kg), continuously administered for 15 days. Pulmonary arterial pressure(PAP), serum erythropoietin(EPO), C-reactive protein(CRP) and growth-promoting factor(IGF-1) levels were measured in every group. The pathological changes of heart tissue were observed and the superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) activity and malondialdehyde(MDA) level were determined. Results Compared with the plain group, the PAP of the HAPH model group, the positive group, the alcohol extract of TBR group, and the water extract of TBR group rats were significantly increased, and the serum CRP and EPO levels were increased. The activity of SOD and GSH-Px in myocardium was weakened, and the content of MDA was increased. The serum levels of IGF-1 in plateau pulmonary hypertension group, alcohol extract of TBR group and water extract of TBR group rats were increased. The serum levels of IGF-1 in the positive group rats were decreased, and the difference was statistically significant(P<0.05). Compared with the HAPH group, the PAP of the positive group and alcohol extract of TBR group rats decreased, The serum levels of CRP and EPO in the positive group, alcohol extract of TBR group and water extract of TBR group rats decreased. The serum levels of IGF-1 in alcohol extract of TBR group and water extract of TBR group rats were increased. The activities of SOD and GSH-Px in heart tissues were decreased, and the content of MDA was increased. The serum IGF-1 levels in the positive group were decreased, and the difference was statistically significant(P<0.05). Conclusion The extract of Terminalia billerica Roxb.
