rhIGF-1/BP3对新生幼鼠支气管肺发育不良的预防作用

摘要 目的探讨重组人胰岛素样生长因子结合蛋白-3(rhIGF-1/BP3)对新生幼鼠支气管肺发育不良(BPD)的预防作用。方法35只SPF级怀孕Wistar大鼠,10只喂养22 d后剖宫产娩出幼鼠作为对照组(腹腔注射生理盐水1 mg/kg),其余25只大鼠建模成功获得20只BPD模型幼鼠均分为BPD模型组(腹腔注射生理盐水1 mg/kg)及干预组(给予rhIGF-1/BP31 mg/kg);比较干预2周后3组新生幼鼠的体质量及生存率,比较干预前后3组新生幼鼠的肺功能[呼气阻力(Re)、吸气阻力(Ri)、最大通气量(MVV)、肺顺应性(C1)、第0.3秒用力呼气容积占用力肺活量的比值(FEV0.3/FVC)]和肺动脉平均压、心输出量、右心室壁厚度、右心室腔大小,比较干预2周后3组新生幼鼠的肺泡计数水平、肺泡平均截距及肺组织胰岛素样生长因子-1(IGF-1)蛋白相对表达量。结果干预后,模型组新生幼鼠体质量、生存率、MVV、C1、FEV0.3/FVC、心输出量、肺泡计数水平、IGF-1蛋白相对表达量较对照组降低,Re、Ri、肺动脉平均压、右心室壁厚度、右心室腔大小、肺泡平均截距水平较对照组升高(P<0.05);干预组新生幼鼠体质量、生存率、MVV、C1、FEV0.3/FVC、心输出量、肺泡计数水平、IGF-1蛋白相对表达量较模型组升高,Re、Ri、肺动脉平均压、右心室壁厚度、右心室腔大小、肺泡平均截距水平较模型组降低(P<0.05)。结论腹腔注射rhIGF-1/BP3干预后,BPD新生幼鼠生长情况、肺功能、心功能均得到改善,肺功脉平均压降低,其机制可能与IGF-1表达量有关。 Objective To investigate the preventive effect of recombinant human insulin-like growth factor-binding protein-3(rhIGF-1/BP3)on bronchopulmonary dysplasia(BPD)in neonatal rats.Methods Thirty-five pregnant SPF-grade Wistar rats were collected in this study,ten of which were allowed to deliver by cesarean section after 22 days and served as the control group(received normal saline 1 mg/kg).The remaining 25 rats were used to establish the BPD model,20 of which were successfully modeled and divided into the BPD model group(n=10,received normal saline 1 mg/kg)and the intervention group(n=10,received rhIGF-1/BP31 mg/kg).After 2-week intervention,the body weight and survival rate of the 3 groups were compared;the lung function(exhalation resistance,inhalation resistance,maximal voluntary ventilation,lung compliance,ratio of forced expiratory volume to forced lung capacity at 0.3 seconds),mean pulmonary arterial pressure,cardiac output,right ventricular wall thickness,and right ventricular cavity size of the 3 groups were compared before and after intervention;alveolar count,mean linear intercept and the relative expression of insulin-like growth factor-1(IGF-1)protein were compared among the 3 groups after 2-week intervention.Results After intervention,compared with the control group,body weight,survival rate,MVV,C1,FEV0.3/FVC,cardiac output,alveolar count level,IGF-1 protein relative expression level were lower in the model group,while Re,Ri,mean pulmonary artery pressure,right ventricular wall thickness,right ventricular cavity size,and mean alveolar intercept level were higher(P<0.05);compared with the model group,body weight,survival rate,MVV,C1,FEV0.3/FVC,cardiac output,alveolar count level and IGF-1 protein relative expression level were higher in the intervention group,while Re,Ri,mean pulmonary artery pressure,right ventricular wall thickness,right ventricular cavity size,and mean alveolar intercept were lower(P<0.05).Conclusion After the intervention of rhIGF-1BP3 in rats with bronchopulmonary dysplasia,the gro
