摘要
目的观察LKB1在胸腺瘤中的表达水平及其影响Thy0517胸腺瘤细胞系增殖、凋亡和侵袭的作用机制。方法应用qRT-PCR技术检测胸腺瘤组织及瘤旁组织中LKB1和mTOR表达水平,在Thy0517细胞中转染pcDNA3.1(+)/LKB1真核表达质粒上调LKB1表达,以MTT检测细胞增殖活力,流式细胞术检测细胞凋亡情况,Transwell检测细胞侵袭能力,Western-blot检测LKB1和mTOR蛋白表达水平。结果LKB1 mRNA在胸腺瘤组织中的表达水平低于癌旁组织(P<0.05);mTOR mRNA在胸腺瘤组织中的表达水平高于癌旁组织(P<0.05)。Thy0517细胞系转染上调LKB1表达后,与对照组与空质粒组比较,实验组的细胞增殖能力受抑制、细胞凋亡率升高、侵袭能力下降、mTOR蛋白表达水平下调(P<0.05)。结论LKB1表达失调与胸腺瘤发病相关,上调LKB1表达可降低mTOR活性,从而引起胸腺瘤细胞的增殖和侵袭能力下降,促进细胞凋亡。
Objective To observe the expression of LKB1 in thymoma and the mechanism of its effect on the proliferation,apoptosis and invasion of thymoma cell line Thy0517.Methods The expression levels of LKB1 and mTOR in thymoma and paracancerous tissue were detected by qRT-PCR.The expression of LKB1 was upregulated by pcDNA3.1(+)/LKB1 transfection in Thy0517,and cell proliferation activity was detected by MTT,apoptosis was detected by flow cytometry,cell invasive ability was detected by Transwell,and the protein expression levels of LKB1 and mTOR were detected by Westernblot.Results The expression level of LKB1 mRNA in thymoma tissue was lower than that in paracancerous tissue(P<0.05);the expression level of mTOR mRNA in thymoma tissue was higher than that in paracancerous tissue(P<0.05).After expression of LKB1 was upregulated by transfection in Thy0517,compared with the control group and the empty plasmid group,the cell proliferation ability was inhibited,the apoptosis rate was increased,the invasive ability was decreased,and the mTOR expression level was decreased in the study group(P<0.05).Conclusion Abnormal expression of LKB1 was associated with the pathogenesis of thymoma.Upregulating the expression of LKB1 can decrease mTOR activity,which can lead to the decrease of proliferation and invasion of thymoma cells and promote apoptosis.
