血红铆钉菇子实体不同极性萃取部位的总酚含量及抗氧化活性 被引量:2

Total Phenolics and Antioxidant Activity of Chroogomphus rutilus Fruiting Body Extracts with Different Polarities

摘要 为考察血红铆钉菇(Chroogomphus rutilus)子实体的抗氧化活性,利用有机溶剂萃取法萃取得到血红铆钉菇子实体石油醚、乙酸乙酯、正丁醇和水4种不同极性萃取部位,采用Folin-Ciocalteu比色法对4种萃取物的总酚含量进行测定;选用DPPH法、ABTS法、FRAP法以及卵黄蛋白脂质过氧化法对4种萃取物进行体外抗氧化活性分析。结果表明:4种萃取物中,多酚提取率从高到低为正丁醇、乙酸乙酯、石油醚、水。在相同浓度条件下,与相同浓度的阳性对照2,6-二叔丁基对甲酚(BHT)相比,1000μg/mL石油醚萃取物,250、500、1000μg/mL乙酸乙酯萃取物,500、1000μg/mL正丁醇萃取物对DPPH自由基的清除能力较高;15.625μg/mL石油醚和正丁醇萃取物对Fe3+的还原能力与相同浓度BHT相比差异不显著;15.625μg/mL石油醚、乙酸乙酯、正丁醇和水4种萃取物,1000μg/mL乙酸乙酯萃取物对ABTS自由基的清除能力与相同浓度的BHT相比差异不显著;1000μg/mL石油醚、正丁醇萃取物,250、500、1000μg/mL乙酸乙酯萃取物抑制蛋白脂质过氧化的能力与相同浓度BHT相比差异不显著。 Antioxidant activities of different solvent extracts from the edible-medicinal mushroom Chroogomphus rutilus were studied in vitro.The extracts were prepared using petroleum ether,ethyl acetate(EtOAC),n-butanol and water,respectively.The total phenolic contents of the four extracts were estimated by the Folin-Ciocalteu colorimetric method.The antioxidant activities of the four fractions were measured by DPPH assay,ABTS assay,ferric reducing antioxidant power(FPAP)assay,and lipid peroxidation inhibition capacity assay,respectively.The results indicated that the total polyphenols extraction rates of the four extracts were stepwise decreased in the order of n-butanol extract,EtoAC extract,petroleum ether extract and water extract.The 1000μg/mL petroleum ether extract,250,500,and 1000μg/mL EtoAC extracts and 500 and 1000μg/mL n-butanol extracts showed significantly higher DPPH scavenging ability than that of the positive control 2,6-di-tert-Butyl-4-methylphenol(BHT)at corresponding concentrations.At 15.625μg/mL,petroleum ether and n-butanol extracts showed the same Fe3+reducing capacity as the control BHT.All the four types of extracts at 15.625μg/mL and the 1000μg/mL EtOAC extract showed the same ABTS+·radical scavenging ability as those of BHT at corresponding concentrations.High concentrations of petroleum ether and n-butanol extracts(1000μg/mL)and EtOAC extracts(250,500,1000μg/mL)showed the same anti-lipid-peroxidation capability as those of BHT at corresponding concentrations.
