雷公藤多苷对类风湿关节炎患者成纤维样滑膜细胞α7nAChR及炎症因子的作用 被引量:9

摘要 目的 :观察雷公藤多苷对类风湿关节炎(RA)患者成纤维样滑膜细胞α7 N型乙酰胆碱受体(α7nAChR)及炎症因子的作用。方法:取行膝关节置换术的RA患者的关节滑膜组织6例,采用酶消化法进行分离培养成成纤维样滑膜细胞(FLS),采用HE染色、免疫组化法鉴定FLS。分为模型组、激动剂组、抑制剂组、西药组、雷公藤多苷组,分别用生理盐水、PNU-282987、α-银环蛇毒素、甲氨蝶呤、雷公藤多苷进行药物干预。结果:①α7nAChR的mRNA表达:与模型组、抑制剂组比较,雷公藤多苷组α7nAChR mRNA表达的差异均有统计学意义(P<0.05)。雷公藤多苷组较激动剂组α7nAChR mRNA的表达减少,差异有统计学意义(P<0.05),雷公藤组较西药组α7nAChR mRNA的表达增加,差异有统计学意义(P<0.05)。②α7nAChR、NF-κBp65、p-STAT3蛋白表达:与模型组、抑制剂组比较,雷公藤多苷组α7nAChR表达均增多,NF-κBp65表达减少;p-STAT3表达均减少,差异均有统计学意义(P<0.05);雷公藤多苷组较激动剂组p-STAT3表达减少,差异有统计学意义(P<0.05);雷公藤多苷组较西药组α7nAChR表达增多,差异有统计学意义(P<0.05)。③炎症因子:雷公藤多苷组IL-17、HMGB1含量较模型组均降低,差异有统计学意义(P<0.05);雷公藤多苷IL-17较激动剂组均升高,差异有统计学意义(P<0.01),较抑制剂组降低(P<0.05);雷公藤多苷组HMGB1水平较抑制剂明显降低,差异有统计学意义(P<0.05)。结论:雷公藤多苷可能通过促进α7nAChR的表达,激活胆碱能抗炎通路,抑制NF-κB信号通路活化及炎症因子的生成,干预RA滑膜炎的发生。 Objective:To observe the effects of tripterygium glycosides on fibroblast-like synoviocyteα7 nicotinic acetylcholine receptor(α7 nAChR) and inflammatory factors in patients with rheumatoidarthritis(RA). Methods:Synovial tissues from 6 RA patients after knee arthroplasty were digested and cultivated into FLS cells. HE staining and immunohistochemistry were conducted. Samples were divided into model group,agonist group,inhibitor group,western medicine group,tripterygium glycosides group,and were treated with saline,PNU-282987,α bungarotoxin,methotrexate,tripterygium glycosides,respectively. Results:(1)Expression of α7 n AChR:The tripterygium glycosides group showed statistical difference from the model group and the inhibitor group(P< 0.05). The expression level of α7 nAChR mRNA in the tripterygium glycosides group was less than that in the agonist group(P< 0.05),and the expression level of α7 nAChR mRNA in the tripterygium glycosides group was higher than that in the western medicine group(P< 0.05).(2)Expression of α7 7 nAChR,NF-κB p65,and p-STAT3:Compared with the model group,inhibitor group,tripterygium wilfordii group showed increase of α7 nAChR expression(P< 0.05),decrease of NF-κB p65 expression(P< 0.05);the expression of p-STAT3 was decreased(P< 0.05);tripterygium glycosides group showed reduced expression of p-STAT3 compared with agonist group(P< 0.05) and also showed increased expression of α7 nAChR compared with western medicine group(P< 0.05).(3)Inflammatory factors:tripterygium glycosides group showed lower expressions of interleukin-17(IL-17) and high mobility group protein1(HMGB1) contents than the model group(P< 0.05);tripterygium glycosides group were higher than agonist group on IL-17 content(P< 0.01),while lower than the inhibitor group(P< 0.05);tripterygium glycosides group were significantly lower than the inhibitor group on HMGB1 levels(P< 0.05).
