摘要
目的比较KV500与叠氮钠作为溶血液中防腐剂对糖化血红蛋白测定的影响。方法用含不同浓度KV500(0.1%、0.05%、0.01%)及叠氮钠的溶血液溶解相同样品,进行糖化血红蛋白测定,并对结果进行比较。将含0.01%KV500的溶血液于2~8℃保存1个月(开瓶)及1年(不开瓶),用其溶解相同样品,进行糖化血红蛋白测定。将4组溶血液于2~8℃保存1个月(开瓶)及1年(不开瓶),采用纯化水微生物限度法检测KV500的抑菌效果。结果含不同浓度KV500与叠氮钠溶血液测定糖化血红蛋白结果差异无统计学意义(P>0.05)。随保存时间的延长,含0.01%KV500溶血液测定糖化血红蛋白结果差异无统计学意义(P均>0.05)。含不同浓度KV500的溶血液于2~8℃保存1个月及1年后均具有良好抑菌效果,与叠氮钠效果相近。结论KV500可替代叠氮钠作为溶血液的防腐剂,不影响糖化血红蛋白的测定。
Objective To compare the effects of KV500 and sodium azide as preservatives in hemolytic agent on determination of glycosylated hemoglobin.Methods The same samples were dissolved with hemolytic agents containing KV500 and sodium azide,at various concentrations,and determined for glycosylated hemoglobin,and the results were compared.The hemolytic agent containing 0.01%KV500 was stored in opening container at 2~8℃for one and 12 months,with which the same samples were dissolved and determined for glycosylated hemoglobin.Four kinds of hemolytic agents were stored in opening containers at 2~8℃for one and in closed containers for 12 months,based on which the bacteriostatic effect of KV500 was evaluated by purified water microbial limit test.Results No significant difference was observed between the determination results of glycosylated hemoglobin in the samples dissolved with hemolytic agents containing various concentrations of KV500 and sodium azide(P>0.05).The determination result of samples dissolved with hemolytic agent containing 0.01%KV500 showed on significant change with the increasing time for storage(each P>0.05).After storage at 2~8℃for one and 12 months,the hemolytic agents containing various concentrations of KV500 showed similar bacteriostatic effect to that of sodium azide.Conclusion KV500 may be used as a preservative instead of sodium azide in hemolytic agent,which shows no influence on the determination of glycosylated hemoglobin.
