摘要
为天麻药用价值及食用价值的评价提供依据,以不同非挥发性有机酸的含量为考察指标,优化天麻中非挥发性有机酸的酯化及萃取条件,采用气相色谱-质谱联用(GC-MS)建立同时检测天麻中非挥发性有机酸(草酸、苹果酸、柠檬酸、棕榈酸、亚油酸、油酸)含量的方法。结果表明:最佳酯化及萃取条件为10%浓硫酸和5%对甲苯磺酸试剂8mL,于50℃下对样品酯化45min,冷却后加20mL正己烷,超声萃取10min。在优化条件下,天麻中草酸、苹果酸、柠檬酸、棕榈酸、亚油酸和油酸6种非挥发性有机酸的线性关系良好(r^2>0.999),回收率较高(89.2%~102.1%),重现性较好(RSD<4.0%)。天麻中含有丰富的非挥发性有机酸,其中苹果酸的含量较高,平均为5.00mg/g;柠檬酸含量次之,平均为3.47mg/g;油酸含量较低,平均为0.13mg/g。
In order to provide a basis for evaluating medicinal value and edible value of Gastrodia elata,the content of different non-volatile organic acids was used as an indicator to optimize the esterification and extraction conditions of non-volatile organic acids in G.elata,gas chromatography-mass spectrometry(GC-MS)was established to simultaneously detect contents of non-volatile organic acids(oxalic acid,malic acid,citric acid,palmitic acid,linoleic acid,oleic acid)in G.elata.Results:The best esterification and extraction conditions included 10% concentrated sulfuric acid and 5% p-toluenesulfonic acid reagent 8 mL,the sample was esterified at 50℃for 45 min,cooled and added with n-hexane 20 mL,and ultrasonic extraction for 10 min.Under optimized conditions,the linear relationships among oxalic acid,malic acid,citric acid,palmitic acid,linoleic acid and oleic acid in G.elata was good(r^2>0.999),and the recovery rate was greater(89.2%-102.1%)with good reproducibility(RSD<4.0%).G.elata was rich in non-volatile organic acids,of which the content of malic acid was greater,the average was5.00 mg/g,followed by citric acid content of 3.47 mg/g on average,oleic acid was of a lower average of0.13 mg/g.
