摘要
目的研究蒙药清肝九味散对肝纤维化大鼠的作用及其机制。方法大鼠腹腔注射四氯化碳建立肝纤维化大鼠模型。按照体重将SD大鼠随机分为5组:正常组、模型组、对照组和低、高2个剂量实验组,每组10只。正常组与模型组给予蒸馏水10 m L·kg^-1灌胃;对照组灌服还原型谷胱甘肽(GSH)300mg·kg^-1;低、高2个剂量实验组灌服清肝九味散水煎液0.52,4.72 g·kg^-1,1次/天,连续12周。用酶联免疫吸附法检测基质金属蛋白酶1(MMP-1)及基质金属蛋白酶抑制剂1(TIMP-1)的表达水平,用全自动生化分析仪测定血清谷丙转氨酶(GPT)、谷草转氨酶(GOT)和谷氨酰转移酶(γ-GGT)水平。结果模型组、正常组、对照组和低、高2个剂量实验组血清GPT含量分别为(531.82±84.59),(32.32±7.51),(316.98±92.60),(227.59±19.61)和(180.26±49.62)U·L^-1;上述这5组的GOT含量分别为(346.95±61.56),(62.65±13.15),(249.24±63.22),(230.15±78.43)和(115.28±34.25)U·L^-1;上述这5组的γ-GGT含量分别为(120.26±18.54),(25.19±7.76),(68.44±32.45),(77.30±15.12),(55.58±7.44)U·L^-1;上述这5组的MMP-1表达分别为(0.76±3.26),(3.32±1.51),(1.02±2.47),(1.88±3.81)和(2.45±1.64)μg·L^-1;上述这5组的TIMP-1的表达分别为(29.95±15.24),(4.28±3.25),(22.35±5.78),(16.95±15.21)和(9.95±15.44)μg·L^-1;上述这5组的肝组织MMP-1阳性表达面积分别为4.23%,16.15%,8.63%,9.82%和11.17%;上述这5组的TIMP-1阳性表达面积分别为23.93%,6.21%,17.58%,14.80%和10.93%;上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.01)。结论清肝九味散可缓解四氯化碳所致的大鼠肝纤维化,其作用机制可能与下调血清转氨酶水平及调节肝组织MMPs/TIMPs体系有关。
Objective To observe the effects of traditional Mongolian medicine Qingganjiuwei powder on the liver fibrosis in rat and possible mechanisms.Methods The rat was injected with 40%of carbon tetrachloride(CCl4)olive oil solution intraperitoneally to induce liver fibrosis.Rats were divided into five groups:normal group,model group,control group,experimental-L and experimental-H groups,each group had ten rats.Rats in normal group and model group were intragastrically administered distilled water(10 m L·kg^-1 per day);rats in control group were intragastrically administered glutathione at the dose of 300 mg·kg^-1 per day,while rats in the experimental-L and experimental-H group wereintragastrically administered Qingganjiuwei powder(0.52,4.72 g·kg^-1 per day),respectively;for consecutive 12 weeks.The levels of serum glutamic-pyruvic transaminase(GPT),glutamic-oxaloacetic transaminase(GOT)and Gamma-glutamyltransferase(γ-GGT)were determined by automatic biochemical analyzer.The level of matrix metalloproteinase 1(MMP-1)and tissue inhibitor of metalloproteinase 1(TIMP-1)were determined by enzyme linked immunosorbent assay and immunohistochemistry.Results The active of serum GPT in model,normal,control,experimental-L and experimental-H groups were(531.82±84.59),(32.32±7.51),(316.98±92.60),(227.59±19.61),(180.26±49.62)U·L^-1,respectively;the GOT in the above five groups were(346.95±61.56),(62.65±13.15),(249.24±63.22),(230.15±78.43),(115.28±34.25)U·L^-1,respectively;theγ-GGT in the above five groups were(120.26±18.54),(25.19±7.76),(68.44±32.45),(77.30±15.12),(55.58±7.44)U·L^-1,respectively;the MMP-1 in the above five groups were(0.76±3.26),(3.32±1.51),(1.02±2.47),(1.88±3.81),(2.45±1.64)μg·L^-1,respectively;the TIMP-1 in the above five groups were(29.95±15.24),(4.28±3.25),(22.35±5.78),(16.95±15.21),(9.95±15.44)μg·L^-1,respectively;the positive area of MMP-1 protein of liver tissue in the above five groups were 4.23%,16.15%,8.63%,9.82%,11.17%;the positive area of TIMP-1 protein in the above
