广西HBV相关肝细胞癌及其癌旁组织中HBV S基因突变分析 被引量:2

Analysis of HBV S gene mutation in HBV-related hepatocellular carcinoma tissues and pericarcinoma liver tissues in Guangxi

摘要 目的了解广西乙型肝炎病毒(hepatitis B virus, HBV)相关肝细胞癌(hepatocellular carcinoma,HCC)组织及其癌旁组织中HBV S基因突变分布状况。方法收集2015-2016年于广西医科大学附属肿瘤医院手术切除的43例HBV相关HCC患者癌组织及其癌旁组织。采用巢式PCR扩增HBV S基因片段并测序,Chromas和DNAMAN软件分析S基因序列。结果肝细胞癌患者HBV S基因热点突变在癌组织及癌旁组织中的分布差异无统计学意义(P>0.05);小分子蛋白(small hepatitis B surface antigen, SHBs)终止突变只存在于癌组织中,突变位点分别为sC69^*、s W74^*、sL94^*、sW172^*和sW182^*。结论广西HBV相关HCC癌组织及其癌旁组织HBV S基因热点突变分布无特异性,SHBs终止突变可能影响HCC发生发展。 Objective To investigate the distribution of hepatitis B virus(HBV) S gene mutation in hepatocellular carcinoma(HCC)and pericarcinoma liver tissues of Guangxi. Methods Totals of 43 patients with HBV-related HCC who underwent HCC surgery in the Guangxi Medical University Cancer Hospital,were selected as subjects from 2015 to 2016,and the HCC tissues and pericarcinoma liver tissues were collected. The HBV S gene fragments were amplified by nest PCR and sequenced. The sequence of S gene was analyzed by Chromas and DNAMAN software. Results There was no significant difference in the distribution of hot spot mutation of HBV S gene between HCC tissues and pericarcinoma liver tissues(P >0.05). The stop mutation of small hepatitis B surface antigen(SHBs) were only found in cancer tissues,which were sC69^*,sW74^*,sL94^*,sW172^* and sW182^*. Conclusions For HBVrelated HCC in Guangxi,there is no specific distribution of hot spot mutations in HBV S gene between cancer tissues and paracancerous liver tissues,SHBs stop mutation may affect the occurrence and development of HCC.
