溃结宁膏穴位敷贴对溃疡性结肠炎大鼠结肠组织黏膜屏障的作用 被引量:4

摘要 目的观察溃结宁膏穴位敷贴对溃疡性结肠炎大鼠结肠黏膜屏障的作用,探讨其治疗溃疡性结肠炎的作用机制。方法选用SD大鼠40只,随机分为空白组、模型组、敷贴组、SASP组,并建立溃疡性结肠炎大鼠模型造模后敷贴组给予溃结宁膏穴位敷贴,SASP组予SASP灌胃;观察结肠组织大体形态及结肠黏膜损伤程度,光学显微镜下观察结肠组织病理改变,并进行结肠黏膜损伤指数(CMDI)、结肠组织学损伤指数(TDI)评分,同时分别运用ELISA方法测定血清中D-乳酸(D-LA)和二胺氧化酶(DAO)的含量、Western blotting方法检测各组大鼠结肠黏膜组织ZO-1、SOCS1的蛋白表达。结果形态组织学检测:在CMDI、TDI评分上,模型组较空白组升高(P<0.01);敷贴组和SASP组较模型组降低(P<0.05);敷贴组与SASP组比较差异无统计学意义(P>0.05)。ELISA检测:与空白组比较,其余3组的D-LA和DAO含量上升(P<0.05);与模型组比较,敷贴组和SASP组D-LA、DAO差异明显(P<0.05);与SASP组比较,敷贴组各项指标差异不明显(P>0.05)。Western blotting检测:与空白组比较,模型组ZO-1、SOCS1蛋白的表达下降(P<0.01);与模型组比较,敷贴组和SASP组ZO-1、SOCS1明显升高(P<0.05);与SASP组比较,敷贴组各项指标差异不明显(P>0.05)。结论溃结宁膏穴位敷贴可以一定程度上提高模型大鼠ZO-1、SOCS1等蛋白的水平,同时降低血清中D-LA和DAO的含量,从而达到减轻肠道炎症反应,促进结肠黏膜愈合,重建肠黏膜机械屏障的完整性的目的。 Objective:To observe the effect of Kuijiening Ointment acupoint application on colonic mucosal barrier in rats with ulcerative colitis,and to explore its specific mechanism.Methods:40 SD rats were randomly divided into the blank group,the model group,application group and SASP group.Rat models of ulcerative colitis were established.After the models were established,the treatment group was given Kuijiening Ointment acupoint application and SASP.After the treatment,the general morphology of colon tissue and the degree of colonic mucosal damage were observed.The pathological changes of colon tissue were observed under light microscope;CMDI and TDI were observed.The contents of D-LA and DAO in serum were determined by ELISA.Protein expression of ZO-1 and SOCS1 of colonic mucosa were detected by Western blotting.Results:1)Morphological histological examination:In CMDI and TDI scores,the model group was higher than the blank group,with statistical significance(P<0.01).Compared with the model group,the application group and SASP group were lower,with statistical significance(P<0.05).2)ELISA test:Compared with the blank group,the levels of D-LA and DAO in the other three groups increased.3)WB detection:Compared with the blank group,the expression of ZO-1,SOCS1 and other proteins in the model group decreased,with statistical significance(P<0.01).Compared with the model group,the application group and SASP group changed correspondingly,with statistical significance(P<0.05);there was no statistical significance between the application group and SASP group(P>0.05).Conclusion:Kuijiening Ointment acupoint application can improve the level of ZO-1,SOCS1 and other proteins in experimental rats to some extent,and reduce the content of D-LA and DAO in serum,so as to alleviate intestinal inflammation,promote the healing of colonic mucosa,and reconstruct the integrity of intestinal mucosal mechanical barrier.
