基于TLR4/NF-κB信号通路探讨防风治疗溃疡性结肠炎大鼠的机制研究 被引量:2

摘要 目的探讨防风对溃疡性结肠炎大鼠Toll样受体4/核因子κB(Toll-like receptors 4/nuclear factor kappa-B,TLR4/NF-κB)信号通路及炎症因子的影响。方法将24只SPF级雄性SD大鼠通过随机数字表法分为正常组、模型组与治疗组,每组各8只;模型组与治疗组大鼠通过5%的2,4,6-三硝基苯磺酸结肠内灌注建立溃疡性结肠炎大鼠模型,正常组大鼠给予等量生理盐水灌肠;造模后治疗组大鼠给予防风水煎液灌胃治疗,模型组及正常组大鼠予等量生理盐水灌胃,连续14d。在造模前及治疗前后观察大鼠一般情况,记录疾病活动指标评分,治疗结束后收集各组大鼠血清、结肠,测量完整结肠长度,结肠组织苏木精-伊红染色,统计病理学评分;酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测血清中炎症因子白细胞介素-6(interleukin-6,IL-6)、白细胞介素-8(interleukin-8,IL-8)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量;蛋白质印迹法(Western blotting)检测TLR4、NF-κB蛋白表达水平。结果与正常组相比,模型组大鼠疾病活动指标评分显著升高,结肠显著缩短,病理评分显著提高,炎症因子IL-6、IL-8、TNF-α含量显著升高,TLR4、NF-κB蛋白相对表达水平显著提高,差异均有统计学意义(P<0.01);经防风治疗后,治疗组大鼠上述情况有所好转,相较于模型组,疾病活动指标评分明显下降,结肠长度明显增长,病理评分显著下降,炎症因子IL-6、IL-8、TNF-α含量明显下降,TLR4、NF-κB蛋白表达水平明显下降,差异均有统计学意义(P<0.01)。结论防风可通过TLR4/NF-κB信号通路减轻肠道炎症,从而治疗大鼠的溃疡性结肠炎。 Objective To explore the effects of saposhnikovia divaricata on Toll-like receptors 4/nuclear factor kappa-B(TLR4/NF-κB)signaling pathway and inflammatory factors in rats with ulcerative colitis.Methods Twenty-four SPF SD male rats were randomly divided into normal group,model group and treatment group by random number table,with 8 rats in each group;The model group and the treatment group were infused with 5%2,4,6-trinitrobenzenesulfonic acid sol into the colon to establish UC rat model,and the normal group rats were given the same amount of normal saline enema.After the model was established,the rats in the treatment group were given saposhnikovia divaricata water decoction by gavage,and the rats in the model group and the normal group were given the same amount of normal saline by gavage for 14 days.The general conditions of rats were observed before modeling and before and after treatment,and the disease activity index score was recorded.After treatment,the serum and colon of rats in each group were collected,and the length of complete colon was measured.Hematoxylin and eosin staining staining of colon tissue,statistical pathological score,the content of inflammatory factors(IL-6,IL-8 and TNF-α)in serum was detected by enzyme linked immunosorbent assay(ELISA),and Western blotting to detect TLR4 and NF-κB protein expression level.Results Compared with the normal group,the disease activity index score of the model group was significantly higher,the colon was significantly shorter,the pathological score was significantly higher,the contents of inflammatory factors IL-6,IL-8 and TNF-αwere significantly increased,and the relative expression level of protein TLR4 and NF-κB were significantly increased,the differences were statistically significant(P<0.01);After saposhnikovia divaricata treatment,the above situation of rats in the treatment group was improved.Compared with the model group,the disease activity index score decreased significantly,the colon length increased significantly,and the pathological score
