力竭对训练小鼠肝线粒体蛋白质组表达的影响

摘要 应用双向凝胶电泳和质谱技术研究训练小鼠力竭运动后的肝线粒体蛋白质组差异表达,探讨力竭对运动训练机体肝线粒体功能的影响.将20只健康雄性ICR小鼠分为训练组(TB)和训练力竭组(TA),分别进行三周游泳训练,休息两日TA组游泳力竭,24 h后取材.提取肝线粒体蛋白质进行2-DE电泳及图谱扫描,PD-quest软件分析图像,质谱鉴定两组间5倍以上差异表达蛋白点.结果显示蛋白质斑点数TB组323±84个和TA组307±106个,TA组和TB组差异表达蛋白点263个,其中2倍以上80个(48个上调和32个下调),5倍以上6个(上下调点各3个).质谱成功鉴定出TA组下调5倍点,为ECHM、ATPD和MMSA.这表明力竭运动使训练机体肝线粒体蛋白质组发生明显差异表达,能量代谢重要酶ECHM、ATPD和MMSA蛋白表达不足,将导致肝线粒体糖、脂代谢及ATP生成障碍,这可能是力竭疲劳和不利于肝脏的重要原因机制. The technologies of two-dimensional gel electrophoresis and mass spectrum were used to study the differential expression on liver mitochondria proteome of trained mice after exhaustive exercise,exploring the effects of exhaustive exercise on liver mitochondrial function.Totally 20 healthy male ICR mice were divided into training group(TB)and exhaustive training group(TA).Two groups were swimming trained for three week.TA group was exhaustive swimming at 48 hour after swimming training.TA and TB group were killed After 24 hours of exhaustive swimming.The total protein of liver was determined by two-dimensional gel electrophoresis,2-DE map were scanned,PD-quest analysis the differential expressed proteins between the two groups.Mass spectrum identification was used in the more than 5 times difference protein points between the two groups.The results showed that there were 323+84 spots in group TB and there were 307±106 spots in group TA on the detected number of protein spots.Between the two groups there were 263 differences protein spots which were 80 points more than 2 times(48 up-regulated and 32 down regulated)and 6 points more than 5 times(3 up-regulated and 3 down-regulated).The 3 down regulated points more than 5 times of TA group were successfully identified by MS.They are ECHM,ATPD and MMSA.These results indicate that exhaustive exercise leads to obvious differential expression of the protein in the liver mitochondria of the training body.The ECHM,ATPD and MMSA that enzymes are related for the energy metabolism were lack of expression.It leads to liver mitochondrial sugar,lipid metabolism and ATP formation disorder,which may be an important mechanism for exhausting fatigue and detrimental to the liver.
