红耳龟睾丸氧化应激损伤及酵母多糖的干预效应 被引量:2

摘要 为探讨氧化应激对红耳龟睾丸氧化损伤的影响及饲料添加酵母多糖对其氧化损伤的缓解,选取体质量为(387.4±49.5)g的健康成体雄性红耳龟32只,随机分成4组,每组2个平行,每个平行4只龟。试验第1 d给3组试验红耳龟进行腹腔一次性注射10%H2O2,对照组注射等量无菌生理盐水。对照组、应激处理组腹注1 d后取样,基础日粮投喂组、酵母多糖添加组(酵母多糖添加量为1200 mg/kg)腹注1 d后,继续饲喂30 d取样。检测睾丸脏器系数、血清性激素含量及抗氧化酶活性,显微观察睾丸组织切片。试验结果显示,腹腔注射10%H2O2损伤了红耳龟睾丸曲细精管生精上皮,次级精母细胞及精子细胞损伤脱落,脏器系数增大,显著降低了过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶活性(P<0.05),丙二醛含量显著升高(P<0.01);氧化应激对血清性激素卵泡刺激素、黄体生成素和睾酮水平影响较小;酵母多糖能提高抗氧化酶活性,清除自由基。综上所述,氧化应激可能直接造成雄性红耳龟生殖器官损伤影响繁殖功能,酵母多糖能提高抗氧化能力,缓解氧化损伤。 Thirty-two healthy adult male red eared turtle(Trachemys scripta elegans)with body weight of(387.4±49.5)g were randomly divided into A(control),B,C and D groups,and each group was dividied into two-subgroups of each four turtles.Groups B,C and D were given a one-time intraperitoneal injection of 10%H2O2,while the control group was given sterile saline following the same method.The testicular organ coefficient,the contents of serum testosterone(T),follicle stimulating hormone(FSH)and luteinizing hormone(LH)and the activity of antioxidant enzyme were measured and testis histology was observed in samples collected from control group A(non-stress treatment of physiological saline),and test group B(stress treatment)1 day after administration,and in samples collected from test group C(stress+basal diet)and test group D(stress+basal diet+added yeast polysaccharide 1200 mg/kg)30 days after administration to investigate the effect of oxidative stress on the testicular oxidative damage of male red eared turtle and the relief of the oxidative damage by adding yeast polysaccharides to the diet.The results showed that intraperitoneal injection of 10%H2O2 harmed the testicular spermatogenesis epithelium of the red eared turtle,and the secondary spermatophore cells and spermatozoon cells were damaged and shed,with increase in viscera coefficient.The activities of catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GSH-PX)were shown to be significantly decreased(P<0.05),and the content of malondialdehyde(MDA)increased(P<0.01),with little effect of oxidative stress on serum sex hormones FSH,LH and T.Yeast polysaccharide led to improve the activity of antioxidant enzyme and scavenge free radicals.In conclusion,oxidative stress may directly cause damage in reproductive organs of male turtles and negatively impact reproductive function and yeast polysaccharide can improve antioxidant capacity and alleviate oxidative damage.
