EPB41L5 mRNA在食管癌组织、细胞中的表达及对食管癌细胞侵袭和转移的影响

Expression of EPB41L5 in esophageal carcinoma tissues and cell lines and its effects on invasion and metastasis of esophageal carcinoma cells

摘要 目的观察红细胞膜蛋白带4.1类似物5(EPB41L5)mRNA在食管癌组织和细胞中的表达变化,及沉默EPB41L5 mRNA表达对食管癌细胞侵袭、转移能力的影响,并探讨相关作用机制。方法采用qRT-PCR法检测食管癌组织、癌旁组织及食管癌细胞(Eca109、EC9706、TE4)、正常人食管鳞状上皮细胞(Het-1A)中的EPB41L5 mRNA。选取EPB41L5 mRNA表达水平最高的食管癌细胞,分为si-NC组和si-EPB41L组,分别转染si-NC和si-EPB41L。转染48 h后,分别采用Boyden实验、Transwell小室实验检测细胞侵袭能力、迁移能力,采用Western blotting法检测细胞中的上皮-间质转化(EMT)相关蛋白E-cadherin、N-cadherin、Vimentin。结果食管癌组织中EPB41L5 mRNA相对表达量高于癌旁组织(P<0.05)。Eca109、EC9706、TE4细胞中EPB41L5 mRNA相对表达量均高于Het-1A细胞(P均<0.05)。选取EPB41L5 mRNA表达水平最高的Eca109细胞用于转染实验。si-EPB41L5组细胞侵袭能力、迁移能力均低于si-NC组(P均<0.05)。si-EPB41L5组上皮细胞标志物E-cadherin蛋白相对表达量高于si-NC组,间质细胞标志物N-cadherin、Vimentin蛋白相对表达量低于si-NC组(P均<0.05)。结论EPB41L5 mRNA在食管癌组织和细胞系中均高表达。沉默EPB41L5 mRNA表达可抑制食管癌细胞的侵袭、迁移能力,作用机制可能与抑制EMT有关。 Objective To investigate the expression of erythrocyte membrane protein band 4.1-like 5(EPB41L5)in the esophageal carcinoma tissues and cell lines,and to investigate the effects of EPB41L5 on invasion and metastasis of esophageal carcinoma,and to explore the relevant mechanism.Methods The expression of EPB41L5 mRNA in the esophageal cancer tissues,adjacent normal tissues,and esophageal cancer cell lines(Eca109,EC9706,and TE4)and normal human esophageal squamous cells(Het-1A)was detected by qRT-PCR.The esophageal cancer cells with the highest expression level of EPB41L5 were selected,and they were divided into the si-NC group and si-EPB41L5 group,which were transfected with si-NC and si-EPB41L5,respectively.After 48 h of transfection,the cell invasion and migration abilities were detected by Boyden assay and Transwell chamber assay.The epithelial-mesenchymal transition(EMT)-related proteins E-cadherin,N-cadherin,and Vimentin were detected by Western blotting.Results The expression level of EPB41L5 mRNA in the esophageal carcinoma tissues was higher than that in the adjacent tissues(P<0.05).The expression level of EPB41L5 mRNA in esophageal cancer cell lines(Eca109,EC9706,and TE4)was higher than that in the normal human esophageal squamous cells(Het-1A)(P<0.05).Eca109 cells with the highest expression level of EPB41L5 mRNA were selected for transfection experiment.The cell invasion and migration abilities of the si-EPB41L5 group were lower than that of the si-NC group(both P<0.05).The relative expression of E-cadherin in the si-EPB41L5 group was higher than that in the si-NC group,the relative expression of N-cadherin and vimentin protein in interstitial cells was lower than that in the si-NC group(all P<0.05).Conclusions EPB41L5 is highly expressed in esophageal cancer tissues and cell lines.The expression of silencing EPB41L5 may inhibit the invasion and metastasis of esophageal carcinoma by regulating EMT.
