摘要 目的通过对比分析RASSF10基因在肾细胞癌发生和发展过程中的作用,为肾细胞癌的治理提供理论依据。方法选取78例肾细胞癌患者作为研究对象,采用反转录聚合酶链反应检测患者肾细胞癌、癌旁组织和正常组织中RASSF10基因mRNA表达量,采用甲基特异性聚合酶链反应检测患者肾细胞癌、癌旁组织和正常组织中RASSF10基因的甲基化状态,观察肾细胞癌患者病变组织中RASSF10基因水平、甲基化状态与患者临床症状等的关系。结果 RASSF10 mRNA在癌变组织中表达水平低于癌旁组织和正常组织,差异有统计学意义(P <0.05),但癌旁组织和正常组织中的表达量差异无统计学意义(P> 0.05);病变组织甲基化率高达70.5%,显著高于对应的癌旁组织甲基化率(35.9%)和正常组织甲基化率(24.4%),差异有统计学意义(P <0.001); RASSF10在患者肾细胞癌组织中甲基化率高于癌旁组织甲基化率和正常组织甲基化率,差异有统计学意义(P <0.05),癌旁组织甲基化率稍高于正常组织甲基化率,差异无统计学意义(P> 0.05);肾细胞癌组织中甲基化结果、RASSF10基因mRNA水平与患者性别、年龄、肿瘤直径、病理类型和分化程度无相关性(P> 0.05),与临床分期显著相关(P <0.05);肾细胞癌组织甲基化mRNA水平低于未甲基化mRNA水平,差异有统计学意义(P <0.05)。结论 RASSF10基因表达水平、甲基化状态与患者肿瘤分期密切相关,在一定程度上能反映患者RASSF10基因低表达水平可能参与了肾细胞癌的发生发展,为诊断和治疗肾细胞癌提供依据,同时,为广大医护人员积累了经验,可广泛推广。 Objective The role and development process RASSF10 gene in renal cell carcinoma,provide a theoretical basis for the treatment of renal cell carcinoma by comparative analysis. Methods 78 patients with renal cell carcinoma were selected as subjects.Reverse transcription polymerase chain reaction was used to detect the expression of RASSF10 mRNA in renal cell carcinoma,paracancerous tissues and normal tissues.Methylation-specific polymerase chain reaction was used to detect renal tissue.cell carcinoma,adjacent tissues and normal tissues RASSF10 methylation status of the gene,observe the relationship RASSF10 gene level,the methylation status of patients with clinical symptoms such as lesions in patients with renal cell carcinoma. Results The expression level of RASSF10 mRNA in cancerous tissues was lower than that in adjacent tissues and normal tissues ( P <0.05),but there was no significant difference in the expression of RASSF10 mRNA in adjacent tissues and normal tissues ( P >0.05).The methylation rate of lesions was as high as 70.5%,which was significantly higher than the corresponding methylation rate (35.9%) and normal tissue methylation rate (24.4%).The difference was statistically significant ( P < 0.001 );RASSF10 The methylation rate in patients with renal cell carcinoma was higher than that in adjacent tissues and the methylation rate of normal tissues.The difference was statistically significant ( P <0.05).The methylation rate of adjacent tissues was slightly higher than normal.There was no significant difference in tissue methylation rate ( P >0.05).There was no correlation between methylation results and RASSF10 gene mRNA levels in patients with renal cell carcinoma,gender,age,tumor diameter,pathological type and degree of differentiation.There was no statistical significance ( P >0.05),which was significantly correlated with clinical stage ( P <0.05).The methylation mRNA level of renal cell carcinoma was lower than that of unmethylated mRNA ( P <0.05).
