丹参酮ⅡA对慢性肾衰大鼠肾病理损伤及肾间质纤维化的影响 被引量:23

Effect of tanshinone ⅡA on the renal pathological damage and renal interstitial fibrosis of chronic renal failure rats

摘要 目的 研究丹参酮ⅡA(TanⅡA)对慢性肾衰大鼠肾病理损伤及肾间质纤维化的影响。方法 将30只健康雄性SD大鼠随机分为对照组、模型组及实验组,各10只。模型组及实验组连续3周灌胃250 mg·kg^-1腺嘌呤溶液构建慢性肾衰大鼠模型,对照组则灌胃等量生理盐水,建模成功后,实验组腹腔注射15. 0mg·kg^-1TanⅡA,对照组及模型组腹腔注射等量生理盐水,连续干预4周。记录各组大鼠24 h尿量及24 h尿蛋白含量,检测大鼠血清肾功能指标及肾间质纤维化指标含量;以苏木精-伊红(HE)染色及Masson染色观察大鼠肾组织病理学形态及肾间质纤维化损伤程度。结果 对照组、模型组及实验组大鼠血清胶原纤维Ⅳ(C-Ⅳ)含量分别为(11. 96±3. 67),(78. 71±10. 48),(35. 62±8. 51)ng·m L^-1,Ⅲ型前胶原蛋白(PC-Ⅲ)含量分别为(21. 38±6. 22),(66. 92±7. 94),(37. 85±8. 36) ng·m L^-1,层粘连蛋白(LN)含量分别为(28. 19±6. 59),(123. 67±15. 98),(90. 01±12. 07) ng·m L^-1,纤维连接蛋白(FN)含量分别为(155. 38±17. 32),(72. 09±9. 98),(119. 89±14. 36)mg·L^-1,肾组织病理学评分分别为(0. 65±0. 13),(4. 12±1. 47),(1. 07±0. 38)分,肾间质纤维化面积百分比分别为(3. 01±1. 45)%,(69. 49±5. 23)%,(14. 57±2. 04)%,实验组和对照组与模型组比较,差异均有统计学意义(均P <0. 05)。结论 TanⅡA可有效改善慢性肾衰大鼠肾病理损伤,延缓肾间质纤维化进展,改善大鼠肾功能。 Objective To explore the effect of tanshinone ⅡA(Tan ⅡA)on the renal pathological damage and renal interstitial fibrosis of chronic renal failure rats.Methods Thirty healthy male SD rats were divided randomly into control group,model group and test group,10 cases in each group,model group and test group were built chronic renal failure rat model by intragastricly administrated 250 mg·kg^-1 adenine solution for 3 weeks continuously,control group was intragastricly administrated equal amount of normal saline,after the success of the modeling,test group intraperitoneal injected 15.0 mg·kg^-1 Tan ⅡA,control group and model group intraperitoneal injected equal amount of normal saline,1 time·d^-1,intervened continuously for 4 weeks.The 24 h urine volume and 24 h urine protein content of rats in each group were recorded,the serum renal function index and renal interstitial fibrosis index contents were detected;the renal pathology morphology and renal interstitial fibrosis damage degree were observed by hematoxylin and eosin(HE)staining and Masson staining respectively.Results The serum collagen Ⅳ(C-Ⅳ)contents in control group,model group and test group were(11.96±3.67),(78.71±10.48),(35.62±8.51)ng·mL^-1,procollagen Ⅲ(PC-Ⅲ)contents were(21.38±6.22),(66.92±7.94),(37.85±8.36)ng·mL^-1,laminin(LN)contents were(28.19±6.59),(123.67±15.98),(90.01±12.07)ng·mL^-1,fibronectin(FN)contents were(155.38±17.32),(72.09±9.98),(119.89±14.36)mg·L^-1,renal histopathology scores were(0.65±0.13),(4.12±1.47),(1.07±0.38)scores,percentage of renal interstitial fibrosis area were(3.01±1.45)%,(69.49±5.23)%,(14.57±2.04)%.There were significant differences between control group,test group and model group(all P<0.05).Conclusion Tan ⅡA can improve the renal pathological damage of chronic renal failure rats effectively,delay the renal interstitial fibrosis progression and improve the renal function of rats.
