自发性高血压大鼠心肌组织骨形成蛋白和激活素的跨膜抑制剂、转化生长因子β1表达情况及其与心房颤动易感性的关系研究 被引量:2

Expressions of BAMBI and TGF-β1 in Myocardial Tissue and Their Relations to Susceptibility of Atrial Fibrillation in Spontaneously Hypertensive Rats

摘要 背景心肌组织骨形成蛋白和激活素的跨膜抑制剂(BAMBI)可负反馈调节转化生长因子β1(TGF-β1)的表达,具有抗纤维化作用,因此探讨BAMBI、TGF-β1表达情况及其与心房颤动易感性的关系对高血压并心房颤动的防治具有重要意义。目的分析自发性高血压大鼠心肌组织BAMBI、TGF-β1表达情况及其与心房颤动易感性的关系。方法2018年8月-2019年4月,选取8只14周龄清洁级Wistar大鼠作为对照组,8只14周龄清洁级自发性高血压大鼠作为观察组;两组大鼠均饲养至22周龄。比较两组大鼠体质量、收缩压(SBP)、心率、左心结构指标〔包括左心房前后径、左心室舒张末期内径(LVEDD)及左心室收缩末期内径(LVESD)〕、左心室射血分数(LVEF)及心房颤动诱发成功率;采用Masson染色法观察大鼠心肌纤维化情况,采用免疫组化染色法检测心肌组织BAMBI、TGF-β1相对表达量;自发性高血压大鼠心肌组织BAMBI、TGF-β1相对表达量间的相关性分析采用Pearson相关分析。结果(1)两组大鼠体质量和心率比较,差异无统计学意义(P>0.05);观察组大鼠SBP高于对照组(P<0.05)。(2)观察组大鼠左心房前后径、LVESD大于对照组,LVEF低于对照组(P<0.05);两组大鼠LVEDD比较,差异无统计学意义(P>0.05)。(3)观察组大鼠心房颤动诱发成功率高于对照组(P<0.05)。(4)Masson染色结果显示,观察组大鼠纤维化程度重于对照组。(5)观察组大鼠心肌组织BAMBI、TGF-β1相对表达量高于对照组(P<0.05);Pearson相关分析结果显示,自发性高血压大鼠心肌组织BAMBI、TGF-β1相对表达量间无直线相关关系(r=0.185,P=0.661)。结论22周龄自发性高血压大鼠心肌组织BAMBI和TGF-β1表达上调,其中TGF-β1表达上调可导致心肌纤维化加重、左心房重构,进而增加心房颤动易感性;而BAMBI表达上调可在一定程度上拮抗自发性高血压大鼠因TGF-β1表达上调所致心房颤动易感性增加,可能成为临床防 Background Expression of TGF-β1 can be negative-feedback regulated by BAMBI,thus it is of great significance to investigate the relations of expressions of TGF-β1 and BAMBI(which is with anti-fibrosis effect)to susceptibility of atrial fibrillation.Objective To analyze the expressions of BAMBI and TGF-β1 in myocardial tissue and their relations to susceptibility of atrial fibrillation in spontaneously hypertensive rats(SHR).Methods From August 2018 to April 2019,a total of eight 14-week-old Wistar rats were selected as control group and eight 14-week-old SHR as observation group;rats in the two groups were raised to 22-week-old.Body mass,SBP,heart rate,indicators of left ventricular structure(including anteroposterior diameter of left atrium,LVEDD and LVESD),LVEF and success rate of induction of atrial fibrillation were compared between the two groups;Masson staining method was used to observe the myocardial fibrosis status,immunohistochemistry assay was used to detect the relative expression quantity of BAMBI and TGF-β1 in myocardial tissue;Pearson correlation analysis was used to analyze the correlation between relative expression quantity of BAMBI and TGF-β1 in myocardial tissue in SHR.Results(1)There was no statistically significant difference in body mass or heart rate between the two groups(P>0.05),while SBP in observation group was statistically significantly higher than that in control group(P<0.05).(2)Anteroposterior diameter of left atrium and LVESD in observation group were statistically significantly greater than those in control group,while LVEF in observation group was statistically significantly lower than that in control group(P<0.05);there was no statistically significant difference in LVEDD between the two group(P>0.05).(3)Success rate of induction of atrial fibrillation in observation group was statistically significantly higher than that in control group(P<0.05).(4)Masson staining results showed that,observation group showed worse degree of myocardial fibrosis compared with that in contro
