Optimizing the Extraction Technology of Sennoside A and B from Senna Pod by Orthogonal Test

摘要 探索番泻豆荚中番泻苷A、B的最优提取工艺及其稳定性,为扩大药源奠定基础。以番泻豆荚为原料,高效液相色谱法测定番泻苷A、B总含量为试验指标,采用单因素试验结合L9(34)正交试验法优选番泻豆荚中番泻苷A、B的提取参数,并考察优选提取方法所得提取液中番泻苷A、B在不同pH及温度下的稳定性。番泻豆荚番泻苷A、B最佳提取工艺参数为0.5%碳酸氢钠溶液,料液比1︰15 (g/mL),温度70℃,提取2 h,提取2次,在此条件下番泻苷A、B总提取量可达到21.72 mg·g-1;最优提取工艺提取液中番泻苷A、B在不同pH溶剂的稳定性随温度、时间的变化显著,室温下, 24 h内,提取液番泻苷A、B在pH为7.5时较稳定。优化所得工艺经济,可靠,且室温下该提取液中番泻苷A、B稳定,对番泻中番泻苷资源的充分利用提供技术参考价值。 To study the optimum extraction technology and stability of sennoside A and B from senna pod, in order to lay a foundation for expanding drug source. Using the high performance liquid chromatography(HPLC) determined the content of sennoside A and B from senna pod. Taking the total content of them as investigated index. The extraction parameters of sennoside A and B in Senna pod were optimized by single factor experiment combined with L9(34) orthogonal test, and the stability of sennoside A and B in different pH and temperature was investigated. The optimum extraction parameters of sennoside A and B were 0.5% sodium bicarbonate solution, the ratio of material to liquid was 1︰15(g/m L), the temperature was 70 ℃, and the extraction time was 2 h. Under these conditions, the total extraction amount of sennoside A and B could reach 21.72 mg·g-1. The stability of sennoside A and B in the optimum extraction process varied significantly with the temperature and time of different pH solvents. Sennoside A and B were stable at pH 7.5. The optimized process was economical, reliable and stable at room temperature. It could provide technical reference value for the full utilization of sennoside resource in senna.
