卡托普利与水飞蓟宾联用对大鼠酒精性脂肪肝的作用及抗氧化应激机制 被引量:4

The Effect of Captopril and Silybin on the Alcoholic Fatty Liver and the Mechanism of Antioxidant Stress in Rats

摘要 目的:研究卡托普利与水飞蓟宾联合用药对大鼠酒精性脂肪肝的作用以及其抗氧化应激机制。方法:SD大鼠造模后随机分为模型组、卡托普利13. 5 mg/kg组、水飞蓟宾37. 8 mg/kg组以及联合用药组(卡托普利13. 5 mg/kg+水飞蓟宾37. 8 mg/kg),另设正常对照组,第12周末腹主动脉取血,检测血清ALT、AST,肝组织称重检测肝脏指数,制备10%肝匀浆检测各组大鼠TG、TC、MDA、GSH-Px含量及CYP2E1蛋白表达。结果:模型组大鼠肝脏指数、TG、TC、ALT、AST、MDA含量及CYP2E1蛋白表达量明显高于正常组,GSH-Px含量明显低于正常组;卡托普利组、水飞蓟宾组、联合用药组大鼠的肝脏指数、TG、TC、ALT、AST、MDA含量及CYP2E1蛋白表达明显低于模型组,GSH-Px含量明显高于模型组;联合用药组大鼠肝脏指数、TG、TC、ALT、AST、MDA含量、CYP2E1表达明显低于卡托普利组和水飞蓟宾组,GSH-Px含量明显增高。结论:联合用药组能够改善酒精性脂肪肝且比单独使用卡托普利组、水飞蓟宾组效果好。 Objective: To study the effect and mechanism of captopril and silybin on alcoholic fatty liver (AFL) and the mechanism of antioxidant stress in rats. Methods: SD rats were randomly divided into the control group,the model group,captopril group (13. 5 mg/kg),silybin group (37. 8 mg/kg) and combination drug group (captopril 13. 5 mg/kg + silybin 37. 8 mg/kg). At the end of the 12 th week,all rats were taken blood from aorta abdominals,serum levels of ALT,AST were detected,liver tissues were weighed. 10% liver homogenate was prepared to determine the levels of TG,TC,ALT,AST,MDA,GSH-Px and CYP2E1. Results: Compared with the control group,the liver coefficient and protein expression levels of TG,TC,ALT,AST,MDA and CYP2E1 were significantly increased in the model group,while the content level of GSH-Px was decreased. Compared with the model group,the liver coefficient and expressions of TG,TC,ALT,AST,MDA and CYP2E1 in captopril group,silybin group and combination drug group were significantly decreased,while the content level of GSH-Px was increased.The combination drug showed the best effect in all drug groups. Conclusion: The combination of captopril and silybin has certain effect on the alcoholic fatty liver in rats,and the combination shows better effect than single use of them separately.
