猕猴桃藤水提物对肝纤维化模型大鼠的保护作用及机制研究 被引量:1

Study on Protective Effect and Mechanism of Kiwifruit Vine Aqueous Extract on Hepatic Fibrosis Model Rats

摘要 目的探讨猕猴桃藤水提物对肝纤维化(Hepatic fibrosis,HF)模型大鼠的作用及其机制。方法将SD大鼠随机分为正常组、模型组、水飞蓟宾组(0.03 g·kg^(-1))及猕猴桃藤水提物高、低剂量组(10、5 g·kg^(-1))。采用50%四氯化碳橄榄油溶液(1.5 mL·kg^(-1))腹腔注射复制大鼠肝纤维化模型,每周2次,8周后模型复制成功,各组再给予相应药物灌胃4周。HE、Masson染色观察病理变化情况;比色法检测大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)和血清超氧化物歧化酶(SOD)、丙二醛(MDA)的含量;Western Blot法检测转化生长因子β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)蛋白表达水平;qRT-PCR法检测大鼠肝组织TGF-β1和E-cadherin基因表达水平。结果与正常组比较,模型组的肝纤维化程度明显,胶原容积分数明显增加(P<0.01),大鼠血清ALT、AST、MDA明显升高(P<0.01),SOD明显下降(P<0.01),肝组织α-SMA和N-cadherin蛋白表达上调(P<0.01),TGF-β1基因和蛋白表达上调(P<0.01),E-cadherin蛋白和基因表达下调(P<0.01,P<0.05);与模型组比较,猕猴桃藤水提物高、低剂量组肝纤维化程度减轻,胶原容积分数明显下降(P<0.01,P<0.05),大鼠血清ALT、AST、MDA水平明显降低(P<0.01),SOD水平明显升高(P<0.01),肝组织TGF-β1、α-SMA和N-cadherin蛋白表达下降(P<0.01),E-cadherin蛋白表达升高(P<0.01),猕猴桃藤水提物高剂量组TGF-β1基因表达下降(P<0.05)、E-cadherin基因表达升高(P<0.05)。结论猕猴桃藤水提物具抗肝纤维化作用,其机制可能与干预肝纤维化的氧化应激反应、影响肝纤维化大鼠肝组织TGF-β1、α-SMA、N-cadherin和E-cadherin蛋白表达有关。 Objective To explore the effect of kiwifruit vine water extract on hepatic fibrosis(HF)model rats and its mechanism. Methods SD rats were randomly divided into normal group,model group,silibinin group(0.03 g·kg^(-1)),and high-,low-dose groups of kiwifruit vine water extract(10,5 g·kg^(-1)). The rat HF model was replicated by intraperitoneal injection of 50% carbon tetrachloride solution(dissolved in olive oil,1.5 mL·kg^(-1)),twice a week for8 weeks,and each group was given the corresponding drugs by gavage for 4 weeks. HE and Masson staining were used to observe the pathological changes. Alanine aminotransferase(ALT), aspartate aminotransferase(AST),superoxide dismutase(SOD)and malondialdehyde(MDA)levels in rat serum were detected by colorimetric method.Western blot was used to examine transforming growth factor β1(TGF-β1),α-smooth muscle actin(α-SMA),Ecadherin and N-cadherin protein expression. qRT-PCR was used to detect the expression levels of TGF-β1 and Ecadherin genes in rat liver tissues. Results Compared with the normal group,the degree of HF in the model group was obvious,the collagen volume fraction was significantly increased(P<0.01),ALT,AST and MDA in rat serum were significantly increased(P<0.01), SOD was significantly decreased(P<0.01), α-SMA and N-cadherin protein expression in liver tissue was up-regulated(P<0.01), TGF-β1 gene and protein expression was upregulated(P<0.01),E-cadherin protein and gene expression was down-regulated(P<0.01,P<0.05). Compared with the model group,the kiwifruit vine water extract high-and low-dose groups reduced the degree of HF,the collagen volume fraction decreased significantly(P<0.01,P<0.05),ALT,AST and MDA in rat serum decreased significantly(P<0.01). SOD increased significantly(P<0.01), TGF-β1, α-SMA and N-cadherin protein expression in liver tissue decreased(P<0.01),E-cadherin protein expression increased(P<0.01),The expression of TGF-β1 gene decreased(P<0.05)and the expression of E-cadherin gene increased(P<0.05)in the kiwifruit vine water extract hi
