摘要
利用含50 mg/L潮霉素(Hyg)的MS固体培养基成功筛选出转CmSAMDC基因的拟南芥株系TA1和TB1,并对T 3代转基因植株的耐盐性进行了分析。研究结果表明:转SAMDC基因的拟南芥在含不同浓度NaCl的培养基中发芽率、侧根数均显著高于野生型拟南芥的;转CmSAMDC基因拟南芥幼苗在200 mmol/L NaCl条件下能正常生长,且具有较低的丙二醛(MDA)含量,而野生型拟南芥在相同胁迫条件下明显萎蔫、失绿甚至死亡。说明过量表达SAMDC基因可以显著提高拟南芥的耐盐性。
The transgenic Arabidopsis thaliana strains TA1 and TB1 were successfully screened by solid medium containing 50 mg/L hygromycin(Hyg),and the T 3 transgenic seedlings were used to analyze the salt tolerance.The results showed that the germination rate and lateral roots of transgenic Arabidopsis thaliana with CmSAMDC gene were significantly higher than those of wild plants under different treatments of NaCl.Furthermore,the transgenic Arabidopsis thaliana seedlings with CmSAMDC gene could grow normally under 200 mmol/L NaCl and appeared a low level of malondialdehyde(MDA),while wild-type plants wilted,lost green or even died.These indicated that overexpression of CmSAMDC gene significantly improved the salt tolerance of Arabidopsis thaliana.
