摘要
以农杆菌介导法将水稻几丁质酶基因导入甜瓜品种“雪脆蜜2号”,PCR检测发现目的基因业已导入甜瓜植株,Southern blot杂交分析证明目的基因已整合到转基因甜瓜植株的染色组上,即获得了甜瓜转基因植株;田间抗病的生物学鉴定结果证明转基因植株提高了对白粉病的抗性,同对照比较,抗性提高2~4级,转基因甜瓜植株的其它农艺性状与阴性对照基本一致。结论为通过转基因技术能够筛选和培育出高抗白粉病的甜瓜新品种。
In order to obtain transgenic plants,Agrobacterium tumefaciens-mediated was used to transfer chitinase gene into muskmelon variety‘Xue cui bao-2’.The results of PCR detection and southern blot analysis showed that desired gene had been introduced into muskmelon plants,and the genes had been integrated into the genome of transgenic plants,both results suggested that transgenic muskmelon plants were obtained;The biological assay of disease-resistance in land proved that powdery mildew-resistance of transgenic plants had been enhanced,compared with no-transgenic plants,owdery mildewresistance of transgenic plants increased by 2~4 levels;in addition,other agronomic traits of transgenic palnts were basically the same as negative control plants.In the end,the conclusion that a new muskmelon variety with highly resistance to powdery mildew can be screened and bred by transgene technology was drawed.
