氢氟酸表面改性种植体对骨质疏松大鼠骨结合的影响 被引量:1

Effect of hydrofluoric acid treated implant on osseointegration in osteoporotic rats

摘要 目的探讨喷砂钛种植体经过氢氟酸(hydrofluoric acid,HF)表面改性后对骨质疏松(osteoporosis,OP)大鼠骨结合能力的改善作用。方法40颗纯钛圆柱状种植体,经过喷砂处理后随机分为观察组和对照组各20颗,观察组应用体积分数0.2%HF酸蚀处理90s,对照组不经过酸蚀处理,应用扫描电子显微镜、原子力显微镜检测种植体表面粗糙度,X射线光电子能谱分析种植体表面化学成分。SD大鼠20只,均行双侧卵巢切除术制备OP模型,术后12周,将对照组种植体植入大鼠左侧股骨干骺端髓管,观察组种植体植入右侧股骨干骺端髓管;植入12周处死大鼠取出带种植体的股骨,显微镜下观察种植体周围骨组织以及种植体与骨组织界面情况,显微CT测定骨组织形态计量学参数,推出试验测定种植体生物力学。结果扫描电子显微镜示观察组种植体表面较对照组粗糙;原子力显微镜示观察组表面粗糙度均方根(root mean square,RMS)[(51.85±5.74)nm]、Z范围[(552.44±60.15)nm]、表面积差[(26.87±3.04)%]均较对照组[(15.21±2.12)nm、(291.32±33.77)nm、(7.29±0.81)%]增大(P<0.05);X射线光电子能谱分析结果示观察组种植体表面氧元素[(56.21±5.97)%]、氟元素[(1.88±0.33)%]含量高于对照组[(54.29±5.83)%、0],碳元素含量[(32.39±4.00)%]低于对照组[(36.71±4.46)%](P<0.05),钛元素[(9.52±1.32)%]含量与对照组[(9.00±1.01)%]比较差异无统计学意义(P>0.05)。植入12周,显微镜下对照组种植体表面见少量骨小梁及骨组织,观察组种植体表面骨小梁及骨组织明显增加、围绕种植体周围可见新骨形成;观察组骨体积占总体积百分比[(40.3±5.2)%]、骨结合率[(46.9±5.3)%]、骨小梁数目[(3.8±0.4)mm]、连接密度[(34.2±4.0)mm^3]均较对照组[(22.1±2.3)%、(20.4±2.1)%、(2.5±0.3)mm、(23.6±3.1)mm^3]增高(P<0.05),骨小梁厚度、骨小梁间隙与对照组比较差异无统计学意义(P>0.05);观察组最大推出力[(71.7±10.2)N]和� Objective To investigate the effect of hydrofluoric acid(HF)treated grit-blasted titanium implants on osseointegration in osteoporotic rats.Methods After blasting treatment,40cylindrical titanium implants were equally divided into observation group receiving 0.2%HF for 90sand control group receiving no HF treatment.The topographical surface roughness and chemical changes on implant surface were determined by using scanning electron microscope,atomic force microscope,and X-ray photoemission spectroscope.Twenty SD rats were performed bilateral ovariectomy to prepare osteoporotic models.Control group accepted implants in the left distal femora and observation group accepted implants in the right distal femora 12weeks after ovariectomy.All rats were sacrificed 12weeks after implantation to remove the femora with implants.The bone-implant interface and bone trabecular structure around the implant were observed on microscope,the histomorphometrical indexes were determined by micro-CT,and the biomechanical parameters were measured.Results Scanning electron microscopy showed a severer roughness of the surface of implant in observation group compared with control group.Atomic force microscopy results showed significantly larger fluoride-modification of root mean square((51.85±5.74)nm),Z range((552.44±60.15)nm)and the surface area difference((26.87±3.04)%)in observation group than those in control group((15.21±2.12)nm,(291.32±33.77)nm,(7.29±0.81)%)(P<0.05).X-ray photoemission spectroscopy results showed higher contents of oxygen((56.21±5.97)%)and fluorine((1.88±0.33)%)as well as a lower content of carbon((32.39±4.00)%)in observation group than those in control group((54.29±5.83)%,0,(36.71±4.46)%)(P<0.05),while titanium content showed no significant difference between two groups((9.52±1.32%)vs.(9.00±1.01%)(P>0.05).After 12 weeks of implantation,microscopy showed obvious increased bone trabecular and bone tissues,and formation of new bone around the implant in observation group,while few bone trabecular and bone
