摘要
以木聚糖为唯一碳源,采用刚果红平板显色法从土壤中筛选出一株产木聚糖酶活性较高的菌株,经形态学和16S rDNA序列分析确定其为芽孢杆菌属,命名为Bacillus subtilis.YH169。通过单因素和正交试验优化了培养基配方和发酵条件,结果表明:最佳培养基配方为麸皮5.0%、氯化铵0.3%、Mg2+浓度0.03%,初始pH值为5.0,在37℃条件下培养72 h,Bacillus subtilis.YH169产木聚糖酶活性较高,达24.40 IU/mL。
A strain with high xylanase activity was screened from soil by Congo red plate coloration method with xylan as the sole carbon source.The strain was identified as Bacillus subtilis YH169 by morphology and 16S rDNA sequence analysis.The effects of carbon source,nitrogen source,metalion,initial pH value,temperature and culturing time on the activity of xylanase were investigated by single factor experiment and orthogonal experiment.The results showed that the xylanase activity of Bacillus subtilis YH169 reached 24.40 IU/mL after incubated in bran 5.0%,ammonium chloride 0.3%,initial medium pH 5.0,and Mg^2+0.03%at 37℃for 72 h.
