复方樟柳碱对大鼠缺血微动脉血流灌注和自律运动的影响 被引量:9

Effects of Compound Anisodine on Microvascular Blood Perfusion and Vasomotion of Ischemia Arteriole in Rats

摘要 目的:比较复方樟柳碱(CA)及其组分氢溴酸樟柳碱(703)和盐酸普鲁卡因(PHI)干预大鼠小肠壁微动脉缺血后灌注和自律运动的时效。方法:66只雄性SD大鼠按实验所用药物分为CA组、703组和PHI组,每组18只,以及生理盐水(NS)组12只。先行开腹术暴露小肠壁,选择直径95μm左右肠壁微动脉。局部滴加去甲肾上腺素(NE,0.02%)致肠壁微动脉预收缩至血流停滞后,于上述局部分别滴加0.1ml CA原液、703溶液(0.01%)、PHI溶液(0.1%)以及等量NS。利用活体微循环显微观测系统和激光多普勒微循环血流仪,连续检测NE前和药后即刻、1min、5min、10min、15min时各组微动脉血流灌注水平(BP),以各组自身NE前BP为基线,归一化BP得到各时点相对血流灌注水平(RBP);测量药后出现微动脉自律运动的起始时间(Vt)和血流灌注曲线经快速傅里叶变换获得的自律运动频率(VF)和振幅(VA)。结果:NE前各组基线BP无统计学差异(P>0.05),滴加NE可致局部微动脉收缩、血流停滞。滴加各种药物后1min和5min的RBP以CA组最高,几近NE前水平,显著高于其它三组(P<0.01),PHI组也明显高于703组和NS组(P<0.01);药后10min的RBP各组间无显著差异(P>0.05);药后15min的RBP仍以CA组显著高于其它三组(P<0.05)。各组VF无统计学差异(P>0.05);703组和CA组Vt均明显早于PHI组和NS组(P<0.05,P<0.01);CA组VA显著大于其它三组(P<0.01),PHI组大于703组和NS组(P<0.01)。结论:与703及PHI相比,CA可通过对微动脉快速增加血流灌注和激活自律运动,有效恢复微循环血流,改善组织缺血。 Objective:To investigate the effects of compound anisodine(CA)and its components,anisodine hydrobromide(703)and procaine hydrochloride(PHI)on ischemic microvascular blood perfusion and vasomotion of small intestinal wall arterioles in rats.Method:66 male SD rats were divided into CA,703,PHI(n=18 in each group)and normal saline(NS,n=12)groups according to the treatment.After exposing small intestinal wall,rats were pretreated with 0.02%norepinephrine(NE)at 95μm arterioles to induce local contraction and interdict microvascular blood flow.And then 0.1 ml CA,0.01%703,0.1%PHI and NS were given dropwise respectively.Intravital microscope and Laser Doppler Flowmeter were employed to determine the arteriolar blood perfusion(BP),relative blood perfusion(RBP).Meanwhile,the start time(Vt),amplitude(VA),frequency(VF)of microvascular vasomotion among groups were compared using the fast Fourier transform at before NE,0min,1min,5min,10min and 15min.Results:No differences of BP were found before NE administration.After 0.02%NE synchronizing,RBP of 1,5min in CA group was higher than other groups,nearly to the pre-NE level,and RBP in PHI group was significantly higher than 703 and NS groups respectively.There was no significant difference of RBP after 10min in each group(P>0.05).The RBP after 15min was higher in CA group than in the other three groups(P<0.05).The microvascular vasomotion stimulated in 703 and CA groups was very significantly earlier than that of PHI and NS groups,and there were no significant differences in VF among each group,and vasomotion appeared in low-frequency mode.Additionally,VA of CA group was significantly higher than other groups and VA of PHI group was significantly higher than 703 and NS groups.Conclusion:Compared with 703 and PHI components,CA restores microcirculatory blood flow and improves tissue ischemia,partially through increasing the arteriolar blood perfusion level and activating a low-frequency and high-amplitude microvascular vasomotion.
