RECK、MMP9蛋白在三阴性乳腺癌中的表达及与患者临床特征及预后的关系 被引量:10

Expression of RECK and MMMPP9 proteins in triple negative breast cancer and the relationship with clinical characteristics and prognosis

摘要 目的探讨Kazal基序富含半胱氨酸的逆转诱导蛋白(RECK)、基质金属蛋白酶9(MMP9)在三阴性乳腺癌中的表达情况及与患者临床特征的关系。方法选取43例三阴乳腺癌患者和43例健康体检者,分别取其三阴性乳腺癌组织标本和正常乳腺组织标本,采用免疫组化法检测三阴性乳腺癌组织和正常乳腺组织中RECK、MMP9蛋白的阳性表达情况,并分析其与三阴性乳腺癌患者临床特征和预后的关系。结果三阴性乳腺癌组织中RECK蛋白的阳性表达率为32.56%,明显低于正常乳腺组织的93.02%,MMP9蛋白的阳性表达率为88.37%,明显高于正常乳腺组织的6.98%,差异均有统计学意义(P﹤0.01)。不同TNM分期、组织学分级、淋巴结转移情况三阴性乳腺癌患者三阴性乳腺癌组织中RECK蛋白阳性表达率比较,差异均有统计学意义(P﹤0.05);不同淋巴结转移情况三阴性乳腺癌患者三阴性乳腺癌组织中MMP9蛋白阳性表达率比较,差异有统计学意义(P﹤0.05)。随访30个月时,RECK蛋白阳性表达三阴性乳腺癌患者的无进展生存率为71.4%,高于RECK蛋白阴性表达患者的31.0%,MMP9阳性表达三阴性乳腺癌患者的无进展生存率为36.8%,低于MMP9阴性表达患者的100%,差异均有统计学意义(P﹤0.05)。结论三阴性乳腺癌组织中RECK的表达水平较低,MMP9的表达水平较高,RECK蛋白的表达可能与三阴性乳腺癌患者的TNM分期、组织学分级和淋巴结转移情况有关,MMP9蛋白的表达可能与淋巴结转移情况有关。RECK蛋白阴性表达和MMP9蛋白阳性表达的三阴性乳腺癌患者预后较差。 Objective To investigate the expression of reversion inducing cysteine rich protein with Kazal motifs(RECK)and matrix metalloproteinase 9(MMP9)in triple negative breast cancer,and to investigate their relationship with patients’clinical characteristics.Method Forty-three cases of triple negative breast cancer and 43 cases of normal subjects were included in the study,from whom the triple negative breast cancer tissue and normal breast tissue were collected,and then the expressions of RECK and MMP9 proteins in the triple negative breast cancer tissues and normal breast tissues of each group were detected by immunohistochemical assay,the relationship between the expressions of RECK and MMP9 proteins and the clinicopathological parameters and prognosis of patients was analyzed.Result The positive expression rate of RECK in study group was 32.56%,which was significantly lower than that in the control group at 93.02%,and the positive expression rate of MMP9 in triple negative breast cancer tissues was 88.37%compared to the 6.98%in normal breast tissues,the difference were of statistical significance(P<0.01).The expression of RECK protein was related to TNM stage,histological grade and lymph node metastasis of triple negative breast cancer(P<0.05);while the positive rate of MMP9 protein was associated with lymph node metastasis(P<0.05).Follow-up for 30 months,the progression-free survival(PFS)rate of patients with positive RECK expression among triple negative breast cancer patients was 71.4%,and it was significantly higher than the 31.0%in those with negative RECK expression,furthermore,the PFS rate of triple negative breast cancer patients with positive MMP9 expression was 36.8%,which was lower than the 100%among those with negative MMP9,and the differences were of statistical significance(P<0.05).Conclusion The expression of RECK was low and MMP9 was high in triple negative breast cancer tissues.The expression of RECK is likely to be associated with TNM stage,histological grade and lymph node metastasis,while expr
