摘要
目的:建立同时测定市售茵栀黄颗粒中新绿原酸、绿原酸、京尼平苷、1,3-二咖啡酰奎宁酸、木犀草苷、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸、黄芩苷、滨蒿内酯、汉黄芩素、黄芩素、汉黄芩苷及千层纸素A共14个化学成分含量的高效液相色谱方法。方法:采用Diamonsil C18(200 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,流速1.0mL·min^-1,柱温30℃,检测波长325 nm,进样量20μL。结果:茵栀黄颗粒中上述14个化学成分均可实现较好的分离,在一定的线性范围内呈良好的线性关系(r≥0.999 0)、精密度(RSD<2.5%)、稳定性(RSD<2.5%)和重复性(RSD<2.5%)良好,平均回收率(n=9)在95.0%~105.0%范围内,RSD在0.8%~3.1%范围内。8批样品中上述14个成分的含量范围分别为0.535~0.653、2.228~2.717、5.171~5.833、0.216~0.274、0.618~0.827、0.323~0.452、0.163~0.386、0.511~0.685、68.515~77.164、0.013~0.018、0.206~1.356、0.797~1.915、0.174~0.786、0.035~0.136 mg·g^-1。结论:该方法可用于茵栀黄颗粒的质量控制与评价。
Objective:To establish an HPLC method for simultaneous determination of neochlorogenic acid,chlorogenic acid,geniposide,1,3-dicaffeoylquinic acid,luteoloside,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid,4,5-dicaffeoylquinic acid,baicalin,scoparone,wogonin,baicalein,wogonoside and oroxylin A in Yinzhihuang granules bought from market.Methods:High-performance liquid chromatography was carried out on a Diamonsil C18(200 mm × 4.6 mm,5 μm)with the mobile phase of acetonitrile(A)-0.1%formic acid water solution(B)at a flow rate of 1.0 mL·min^-1,a column temperature of 30 ℃,a detection wavelength of325 nm and the sample size of 20 μL by using gradient elution.Results:Above-mentioned 14 compounds were separated well and showed an excellent linear relationship(r≥0.999 0)with high precision,stability and repeatability(RSD<2.5%).In addition,the average rate(n=9)of the 14 above-mentioned compounds was95.0%-05.0% with the RSD value of 0.8%-3.1%.The contents of the above-mentioned 14 compounds of 8 groups were 0.535-0.653,2.228-2.717,5.171-5.833,0.216-0.274,0.618-0.827,0.323-0.452,0.163-0.386,0.5 11-0.685,68.515-77.164,0.013-0.018,0.206^-1.356,0.797^-1.915,0.174-0.786,0.035-0.136 mg·g^-1,respectively.Conclusion:This method can be used as an efficient method for quality control and evaluation of Yinzhihuang granules.
