贮藏温度及时间对大叶相思花粉活力的影响 被引量:5

The Effect of Storage Temperature and Time of Acacia auriculiformis Pollen Viability

摘要 以大叶相思花粉为研究对象,对其贮藏温度(-20℃,5℃,室温)、贮藏时间(7 d,15 d,30 d,365 d,2 190 d)以及活力检测进行研究。使用花粉离体萌发法,I2-KI染色法以及过氧化物酶染色法对新鲜的和贮藏的大叶相思花粉进行活力检测。结果表明:大叶相思新鲜花粉萌发率最高为96.09%。随着贮藏时间的增加,花粉萌发率、花粉管长度以及花粉管条数均呈现下降趋势。在-20℃下贮藏的大叶相思花粉,其萌发率显著高于在5℃和室温条件下贮藏的花粉的萌发率。在-20℃贮藏365 d后,大叶相思花粉萌发率为53.37%,贮藏2 190 d后,花粉萌发率为12.57%。在5℃和室温下贮藏的大叶相思花粉,随着贮藏时间的增加,萌发率呈下降-上升-下降的趋势。I2-KI染色法和过氧化物酶染色法检测大叶相思花粉活力,无显著差异。大叶相思花粉适当的贮藏和检测方法有利于提高相思育种效率,并具有较高的实际应用价值。 Acacia auriculiformis pollen as research subject, the condition of fresh and storing pollen of Acacia auriculiformis for a long period of time and viability testing were studied. Pollen was stored at-20℃, 5℃ and roo m temperature(25℃~30℃) taking for 7, 15, 30, 365 and 2 190 days of storage. Three methods were used for viability testing, which in vitro germination, I2-KI staining and peroxidase staining. The results showed that the fresh pollen of Acacia auriculiformis presented the highest pollen germination(96.09%). With the increase of storage time, pollen germination percentage, pollen tube length and tube number decreased. The germination percentage of pollen stored at-20℃ was significantly higher than those stored at 5℃ and room temperature. The pollen germination percentage was 53.37% after 365 days of storage at-20℃. The pollen germination percentage was12.57% after 2 190 days of storage at-20℃. With the storage time increasing, the germination percentage of Acacia auriculiformis pollen stored at 5℃ and room temperature showed a downward-rebound-downward trend.Irrespective of storage time, the results of I2-KI staining and peroxidase staining test showed there were no significant difference of Acacia auriculiformis. The appropriate pollen storage methods and viability testing methods of Acacia auriculiformis will improve the efficiency of Acacia breeding and have higher practical application value.
