摘要
目的 :探讨神经再生素 (NRF)对离体培养的 PC12细胞、背根神经节细胞、大脑皮层细胞生长影响的分子生物学机制。方法 :采用细胞体外培养、半定量 RT- PCR、Western blot方法 ,观察和比较 NRF组 (添加 NRF)、NGF组 (添加 NGF)和空白对照组 (基础培养基 )中细胞生存状况及相关基因 m RNA及其蛋白水平的表达变化。结果 :NRF作用于离体培养的 PC12细胞 ,可促使其分化 ;作用于背根神经节细胞 ,GAP- 4 3和 NF- L m RNA表达在不同时间呈不同程度的上调 ;作用于大脑皮层细胞 ,GAP- 4 3和 NF蛋白亦比对照组表达量为高。结论 :NRF具有维持细胞生存和促进神经细胞突起生长的作用 ,并可使神经元 GAP- 4 3和 NF m RNA表达及其蛋白合成上调。
Objective:To investigate the effect of PC12 cells, the cultured dorsal root ganglion cells and cerebral cortex cells with the treatment of the nerve regeneration factor (NRF). Method:Cell culture, semi-quantitative RT-PCR and Western blot were performed to detect the change of mRNA and protein of the GAP-43 and NF genes.Results: As compared with control, the gene expression of GAP-43, NF were significantly up-regulated both in the groups of NRF and NGF, to different extent, in a time-dependent manner.Conclusion: The results indicated that NRF could induce the differentiation of PC12 cells and promote the expression of growth association protein 43 and cell skeleton protein NF to maintain neuronal survival and neurite outgrowth.
