幽门螺杆菌cagA基因多态性与胃癌细胞白细胞介素-22 mRNA表达的关系研究 被引量:2

Correlation between Helicobacter pylori cagA gene polymorphism and the expression of interleukin-22 (IL-22) mRNA in gastric cancer cells

摘要 目的研究幽门螺杆菌(Hp)cagA基因多态性与胃癌AGS细胞、SGC7901细胞白细胞介素22(IL-22)mRNA表达的关系,并探讨HpcagA基因多态性对胃癌AGS细胞、SGC7901细胞IL-22mRNA表达模式的影响。方法37℃、5%CO2孵育箱培养AGS细胞、SGC7901细胞;采用微需氧产气袋培养Hp,用cagA基因东亚型HPK5菌株、西方型HP26695菌株感染AGS细胞及SGC7901细胞0.5h、3h、6h、9h、12h、24h;姬姆萨染色方法评估Hp感染AGS细胞、SGC7901细胞效果;RT-PCR法、ELISA法分别检测上述6个时间段AGS细胞、SGC7901细胞IL-22mRNA相对表达量和IL-22浓度。结果cagA基因东亚型HPK5和西方型HP26695菌株感染AGS细胞0.5h、3h、6h、9h、12h、24h所诱导IL-22mRNA的相对表达差异有统计学意义(P<0.05);东亚型菌株诱导AGS细胞IL-22mRNA表达随时间增加而增加,而西方型菌株诱导AGS细胞IL-22mRNA表达3h、12h出现峰值。cagA基因东亚型和西方型菌株感染SGC7901细胞0.5h、3h、6h、9h、12h、24h所诱导IL-22mRNA的相对表达差异有统计学意义(P<0.05);东亚型和西方型菌株诱导SGC7901细胞IL-22mRNA表达均随时间增加而增加。结论①HpcagA基因东亚型HPK5菌株更易诱导IL-22mRNA及其浓度的表达;②HpcagA基因东亚型HPK5菌株感染AGS细胞诱导IL-22产生呈时间依赖性,西方型菌株感染AGS细胞诱导IL-22产生在3h、12h时出现峰值;③HpcagA基因东亚型菌株和西方型菌株感染SGC7901细胞产生的IL-22均随时间的增加而增加。 Objective To investigate the correlation between Helicobacter pylori(Hp)cagA gene polymorphism and the expression of interleukin-22(IL-22)mRNA in gastric cancer AGS cells and SGC7901 cells,and to investigate theeffects of Hp cagA gene polymorphism on the IL-22 mRNA expression pattern of gastric cancer AGS cells and SGC7901 cells.Methods AGS cells and SGC7901 cells were cultured in a 37℃,5%CO 2 incubator.Hp was cultured in an AnaeroPack-MicroAero,and AGS cells and SGC7901 cells were infected with cagA gene East Asia type HPK5 strain and Western type HP26695 strain for 0.5 h,3 h,6 h,9 h,12 h and 24 h.Giemsa staining method was used to evaluate the effects of Hp infecting AGS cells and SGC7901 cells.RT-PCR and ELISA were used to detect the relative expression of IL-22 mRNA and the concentration of IL-22 in AGS cells and SGC7901 cells in the above 6 time periods.Results The relative expression of IL-22 mRNA which were induced by AGS cells infected by cagA gene East Asian type HPK5 and Western type HP26695 strains at 0.5 h,3 h,6 h,9 h,12 h and 24 h was statistically different(P<0.05).The expression of IL-22 mRNA in AGS cells which was induced by East Asian type strain increased with time extension,while the peak expression of IL-22 mRNA in AGS cells which was induced by the Western type strain occurred at 3 h and 12 h.The relative expression of IL-22 mRNA induced by the East Asian and Western type strains of cagA gene infecting SGC7901 cells at 0.5 h,3 h,6 h,9 h,12 h and 24 h was statistically different(P<0.05).The IL-22 mRNA expression in SGC7901 cells induced by East Asian and Western type strains increased with time extension.Conclusion①East Asian type HPK5 strain of Hp cagA gene is more likely to induce the expression of IL-22 mRNA and its concentration.②The IL-22 production induced by East Asian type HPK5 strain of Hp cagA gene infecting AGS cells presents in a time-dependent manner,while the IL-22 production induced by Western type strain infecting AGS cells peaks at 3 h and 12 h.③The IL-22 produced by H
