广西黑衣壮儿童IL-22基因rs1179251、rs2227485位点多态性研究

摘要 目的探讨广西黑衣壮儿童IL-22基因rs1179251、rs2227485位点的多态性分布特点和不同人群间的多态性差异。方法采取多重单碱基延伸法(SNaPshot)与DNA测序相结合的方法对115例广西黑衣壮健康儿童的IL-22基因rs1179251、rs2227485位点进行基因分型检测,并用统计学方法比较不同性别及人群分布差异。结果广西黑衣壮健康儿童IL-22基因rs1179251和rs2227485分别存在CC、CG、GG和GG、AG、AA三种基因型。rs1179251、rs2227485两位点基因型及等位基因频率在广西黑衣壮健康儿童人群中男女性别间差异无统计学意义(P>0.05)。rs1179251位点基因型及等位基因与人类基因组计划项目(Hap Map)公布的HapMap-YRI、HapMap-MKK均有统计学意义(P<0.05),与HapMap-HCB、HapMap-JPT、HapMap-GIH相比差异无统计学意义(P>0.05)。rs2227485位点各基因型及等位基因分布频率与HapMap-MKK比较均有统计学意义(P<0.05),与HapMap-HCB、HapMap-JPT、HapMap-YRI、HapMap-GIH相比差异无统计学意义(P>0.05)。结论IL-22基因rs1179251、rs2227485基因位点多态性在不同种族和地区间存在差异。 Objective To explore the distribution characteristics of polymorphisms of interleukin-22(IL-22)(rs1179251,rs2227485)in Black-clothes Zhuang children in Guangxi and its comparison with different ethnic groups.Methods Polymorphisms of IL-22(rs1179251 and rs2227485)in 115 Black-clothes Zhuang children were genotyped by SNaPshot technique and DNA sequencing,and distribution differences between genders and groups were analyzed by statistical methods.Results CC、CG、GG and GG、AG、AA genotypes were respectively observed in rs1179251 and rs2227485 of IL-22 gene in Black-clothes Zhuang children in Guangxi population.There was no significant difference between male and female(P>0.05).Significant differences were observed between genotypes and alleles of rs1179251 and HapMap-YRI and HapMap-MKK confirmed by human genome project(HapMap)(P<0.05),but there were no significant differences between genotypes and alleles of rs1179251 and HapMap-HCB,HapMap-JPT,HapMap-GIH population(P>0.05).Significant differences were observed between genotypes and alleles of rs1179251 and HapMap-YRI and HapMap-MKK confirmed by human genome project(HapMap)(P<0.05),but there were no significant difference between genotypes and alleles of rs1179251 and HapMap-HCB,HapMap-JPT,HapMap-GIH population(P>0.05).The distribution frequency of each genotype and allele at rs2227485 site was significantly different from HapMap-MKK(P<0.05),but no significant differences were found between the distribution frequency of each genotype and allele at rs2227485 and HapMap-HCB,HapMap-JPT,HapMap-YRI and hapmap-GIH(P>0.05).Conclusion There were differences in polymorphism of IL-22(rs1179251 and rs2227485)among different races and regions.
