摘要 目的探究miR-128-3p对胶质瘤U87细胞的侵袭、迁移及上皮间充质转化的影响.方法 RT-PCR检测VEGFC与miR-128-3p的mRNA水平;荧光素酶报告实验检测VEGF-C与miR-128-3p的靶向关系;Transwell检测细胞侵袭;划痕实验检测细胞迁移;免疫印迹检测VEGF、 MMP-2、 MMP-9、 E-cadherin、 N-cadherin及Vinmentin表达水平.结果 miR-128-3p与VEGF-C之间存在连续结合片段, miR-128-3p靶向下调VEGF-C,上调VEGF-C促进U87细胞侵袭和迁移, miR-128-3p通过靶向下调VEGF-C抑制U87细胞侵袭和迁移.上调VEGF-C促进U87细胞上皮间充质转化, miR-128-3p通过靶向VEGF-C抑制U87细胞上皮间充质转化.结论 miR-128-3p通过靶向下调VEGFC抑制胶质瘤U87细胞侵袭、迁移及上皮间充质转化. Objective To investigate the effect of miR-128-3p on invasion, migration and ep-ithelial mesenchymal transformation of glioma U87 cells. Methods mRNA levels of VEGFC and miR-128-3p were detected by RT-PCR, the target relationship between VEGFC and mir-128-3p by luciferase assay, cell invasion Transwell, cell migration by scratch test. Immunoblot analysis was used to determine the expression levels of VEGF, MMP-2, MMP-9, E-cadherin, N-cadherin and vinmentin. Results There was a continuous binding fragment between mir-128-3p and VEGF-C. The targeted down-regulation of VEGF-C by Mir-128-3p and targeted up-regulation of vegf-c pro-moted the invasion and migration of U87 cells. Mir-128-3p inhibited the invasion and migration of U87 cells by targeted down-regulation of VEGF-C. U87 cell epithelial mesenchymal transformation was promoted by up-regulation of VEGF-C, while miR-128-3p inhibited epithelial mesenchymal transformation by targeting VEGF-C. Conclusion miR-128-3p inhibits the invasion, migration of U87 cells and epithelial mesenchymal transformation of glioma U87 cells by targeting VEGFC down-regulation.
