DNA甲基转移酶在维吾尔族人群网膜脂肪中的表达及其与脂肪因子的相关性分析

DNMT expression in the omental adipose tissue and its correlation with adipocytokines in Xinjiang Uygur population

摘要 目的检测维吾尔族人群网膜脂肪组织DNA甲基转移酶(DNMTs)的mRNA表达水平,分析DNMTs与脂肪因子的相关性。方法选取维吾尔族体重正常者(Con组)35例及单纯性肥者(OB组)35例,收集一般资料和生化指标。RT-PCR检测网膜脂肪组织DNMTs及脂肪因子mRNA表达水平,Westren blot检测蛋白表达水平。结果与Con组比较,OB组BMI、WC、臀围、WHR更高,差异有统计学意义(P<0. 05)。OB组网膜脂肪组织中DNMT1、DNMT3a、DNMT3b和TNF-αmRNA表达水平高于Con组(P<0. 05),APN mRNA表达水平低于Con组(P<0. 05);TNF-αmRNA表达与DNMT1 mRNA表达水平呈正相关(r=0. 662,P<0. 05),与DNMT3a、DNMT3b mRNA表达不相关(P>0. 05),DNMT1 mRNA表达与APN mRNA表达不相关(P>0. 05)。结论维吾尔族肥胖人群腹部脂肪组织DNMTs表达增加,可能与肥胖诱发的炎症状态有关。 Objective To investigate DNMTs mRNA expression in omental adipose tissue,and to analyze the relationship between DNMTs and adipose tissue factor in Xinjiang Uygur population.Methods The Uyghur subjects were selected and divided into two groups:normal control group(Con group,n=35)and obesity group(OB group,n=35). The mRNA level of DNMTs and the adipocytokines were detected by RT-PCR. Protein expression levels were tested using western blot.ResultsThe level of BMI,WC,HC and WHR were significantly higher in the OB group than in Con group(P<0. 05). The expression level of DNMT1,DNMT3 a,DNMT3 b and TNF-α were significantly higher in the OB group than in Con group(P<0. 05). The mRNA level of APN was significantly lower in the OB group than in Con group(P<0. 05). The expression of TNF-α was positively correlated with DNMT1(P<0. 05),but not with DNMT3 a(P>0. 05)and DNMT3 b(P>0. 05). There was no singnifican correlation between the expression of DNMT and APN(P>0. 05).ConclusionDNMTs expression was increased in obesity subjects in Uygur population,which may be related with obesity induced inflammation.
