摘要
目的探讨腹部脂肪组织APN、TNF-α和MCP-1基因启动子区DNA甲基化及mRNA表达水平与新疆维吾尔族T2DM发生发展的相关性。方法分别选择维吾尔族正常(NC)、肥胖(Ob)和2型糖尿病(T2DM)个体50例、48例和26例。收集腹部脂肪组织,变性高效液相色谱技术(DHPLC)检测上述基因DNA甲基化,实时定量PCR(RTPCR)检测mRNA表达。结果比较NC、Ob及T2DM组间DNA甲基化阳性率,APN逐渐增加(P<0.05),TNF-α逐渐降低(P<0.01);NC组APN mRNA表达显著高于Ob及T2DM组,TNF-α和MCP-1均显著低于Ob及T2DM组(P<0.05)。APN mRNA表达与BMI、DBP、FPG、TG、LDL显著负相关(P<0.05)。TNF-αmRNA表达与BMI、FPG和TG显著正相关(P<0.05);MCP-1 mRNA表达与体质量、BMI和腰围显著正相关(P<0.05)。APN基因DNA甲基化与其mRNA表达显著负相关(P<0.01),与FPG、TC、TG和LDL显著正相关(P<0.05);TNF-α基因DNA甲基化与其mRNA表达、BMI、FPG、TC、TG和LDL显著负相关(P<0.05)。结论 APN、TNF-α和MCP-1在腹部脂肪组织差异表达可能是引起新疆维吾尔族人群肥胖和T2DM的原因;糖脂代谢紊乱可通过导致腹部脂肪组织APN和TNF-α甲基化水平变化而影响其mRNA表达水平。
Objective To and promoter methylation investigate the relationship of type 2 diabetes mellitus(T2DM) with mRNA expression of adiponectin(APN), tumor necrosis factor alpha( TNF-α), monocyte chemotactic cy- tokines protein 1 ( MCP-1 ) gene in abdominal adipose tissues from Xinjiang Uygur population. Methods The ab- dominal adipose tissues of Uyghur subjects were collected and divided into normal control group ( NC, n = 50 ), obesity group( Ob, n =48 )and T2DM group( n = 26). DNA methylation was detected by denaturing high perform-ance liquid chromatography and the mRNA was detected by real time-PCR. Results Compared APN was increased (P 〈0.05 ) ; TNF-α was decreased( P 〈0.01 ). APN in control group was significantly higher than that of obesity and T2DM groups, while mRNA of TNF-α and MCP-1 in control group was significantly higher than that of obesity and T2DM groups (P 〈 0. 05 ). The mRNA expression of APN was significantly negative correlated with BMI, DBP, FPG, TG, LDL(P 〈 0. 05 ) ; The mRNA expression of TNF-a was significantly positive correlated with BMI, FPG, TG(P 〈0. 05); The mRNA expression of MCP-l was significantly positive correlated with body weight, BMI and waist circumference (P 〈 0.05 ). DNA methylation of APN was significantly negative correlated with the mRNA ex- pression level of APN (P 〈 0.01 ) , and was significantly positive correlated with plasma level of FPG, TC, TG and LDL( P 〈0. 05 ) ; DNA methylation of TNF-α was significantly negative con'elated with the mRNA expression level of TNF-α, BMI, FPG, TC, TG and LDL. Conclusions The different expression of APN, TNF-α and MCP- 1 gene in abdominal adipose tissue may be a reason to explain the obesity and T2DM in the Xinjiang Uyghur population; In addition the glucose and lipid metabolic disorder may result in DNA methylation and then affect the mRNA expres-sion of APN and TNF-α.
